Effects Of Short-Term Pesticides Exposure On Swimming Behavior, Tissue Morphology, Oxidative-Nitrative Stress, Antioxidant Expression, And Cellular Apoptosis In Kidneys Of Goldfish, Esmirna Cantú

Theses and Dissertations

The prevalence of pesticides in the aquatic environment continues to increase due to anthropogenic activities and poses a threat to aquatic organisms. The aim of this study was to determine the short-term exposure (one-week) of pesticide mixtures (metolachlor, linuron, isoproturon, tebucanazole, alconifen, atrazine, pendimethalin; azinphos-methyl) on swimming behaviors, kidney morphology, and expression of nitrotyrosine protein (NTP), dinitrophenyl protein (DNP), superoxide dismutase (SOD), catalase (CAT), renin, and cellular apoptosis in goldfish (Carassius auratus). Results indicated that both low- and high-dose pesticide mixtures decreased the distance swimming behavior. Histological analysis showed pesticide exposure alters kidney morphology. DNP, NTP, and renin expressions, and …

Bioaccumulation Of Polychlorinated Biphenyl Compounds And Mercury In A Mining Impacted Aquatic Ecosystem, Michelle Bollini Jan 2023

Bioaccumulation Of Polychlorinated Biphenyl Compounds And Mercury In A Mining Impacted Aquatic Ecosystem, Michelle Bollini

Dissertations, Master's Theses and Master's Reports

The Keweenaw area continues to be influenced by the century of copper mining that ended nearly 50 years ago. This project is focused on Torch Lake, an aquatic ecosystem that has been heavily impacted by mining waste disposal. The watershed has been impaired by mine discharge and tailings, smelter and smokestack plumes, and poor waste disposal practices. The lake is listed as a Great Lakes Area of Concern with beneficial use impairments of restrictions on fish consumption and a degraded benthic community. Polychlorinated biphenyl compounds (PCBs) and methylmercury (MeHg) are persistent, bioaccumulative, and toxic substances (PBTs). These contaminants pose threats …

The Impacts Of Embryonic Arsenic Exposure Of Fundulus Heteroclitus, Torey Bowser Aug 2021

The Impacts Of Embryonic Arsenic Exposure Of Fundulus Heteroclitus, Torey Bowser

Electronic Theses and Dissertations

Arsenic is a toxic metalloid that exceeds safe drinking water standards in groundwater in many locations worldwide. Arsenic exposure in fish has been linked to destruction of gill tissues, impairment of growth, decreased muscle mass, memory impairment, increased aggression, and avoidance behaviors. We examined the behavior of mummichogs (Fundulus heteroclitus) following arsenic exposure during development in two studies. Embryos were collected from fish from three reference sites: Scorton Creek (SC), Massachusetts, Wells Harbor (WE), Maine, and Block Island (BLOC), Rhode Island and two contaminated sites: Callahan Mine (CM), Brooksville, Maine, and New Bedford Harbor (NBH), Massachusetts. Embryos were …

Hepatobiliary Polycyclic Aromatic Hydrocarbons In Pelagic Fishes Of The Gulf Of Mexico, Madison R. Schwaab Oct 2020

Hepatobiliary Polycyclic Aromatic Hydrocarbons In Pelagic Fishes Of The Gulf Of Mexico, Madison R. Schwaab

USF Tampa Graduate Theses and Dissertations

Fisheries populations and their ecosystems are negatively impacted by both chronic and acute inputs of pollutants, including oil spills such as the Deepwater Horizon platform blowout in 2010 in the Gulf of Mexico (GoM). After Deepwater Horizon, toxicological studies of demersal fishes of the northern GoM were undertaken to characterize impacts and to establish baseline contaminant levels in the aftermath of the spill. In this study, I quantify polycyclic aromatic hydrocarbon (PAH) concentrations in eight pelagic fish species to demonstrate oil exposure differences between species, region and time. Analysis of biliary PAH metabolite equivalents using high performance liquid chromatography was …

Polychlorinated Biphenyls, Organochlorine Pesticides, And Polycyclic Aromatic Hydrocarbons In Snapper (Family Lutjanidae) From Cuba And The Wider Gulf Of Mexico, Brigid E. Carr Jun 2020

Polychlorinated Biphenyls, Organochlorine Pesticides, And Polycyclic Aromatic Hydrocarbons In Snapper (Family Lutjanidae) From Cuba And The Wider Gulf Of Mexico, Brigid E. Carr

USF Tampa Graduate Theses and Dissertations

The Snapper species assemblage is economically and environmentally important to all three countries bordering the Gulf of Mexico (GoM). Polycyclic aromatic hydrocarbons (PAHs), organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) were investigated in seven snapper species located in the GoM, with a specific focus on the those caught along the northwestern coast of Cuba. Hepatic samples were processed using QuEChERs extraction methodology and analyzed using gas chromatography with tandem mass spectrometry (GC-MS/MS). Nearly all fish included in this study contained detectable amounts of all three groups of these contaminants, regardless of location, and across all regions of the Gulf, indicating …

Per- And Polyfluoroalkyl Substances In Consumable Fish & Surface Waters Near Sites Using Aqueous Film-Forming Foams: Potential Risks, Suggested Regulatory Standards And Policy Approaches For Massachusetts, Tachalla Gibeau May 2020

Per- And Polyfluoroalkyl Substances In Consumable Fish & Surface Waters Near Sites Using Aqueous Film-Forming Foams: Potential Risks, Suggested Regulatory Standards And Policy Approaches For Massachusetts, Tachalla Gibeau

Sustainability and Social Justice

Per- and polyfluoroalkyl substances (PFAS) are emerging contaminants of concern with potential for adverse effects on both human and environmental health. Hence, long-chain PFAS compounds used in legacy aqueous film-forming foams (AFFFs) are being phased out, resulting in the stockpiling of them at various commercial installations. PFAS contaminated runoff from these facilities has the potential to contaminate nearby surface water bodies and the fish inhabiting them. An analytical approach was used to identify surface water bodies in Massachusetts that are habitats for fish susceptible to PFAS contamination and where said fish are also popularly consumed by locals – thereby representing …

Molecular Responses To Deepwater Horizon Oil Spill Contaminants In Estuarine Fish, Elizabeth Jones Aug 2019

Molecular Responses To Deepwater Horizon Oil Spill Contaminants In Estuarine Fish, Elizabeth Jones

Dissertations

The 2010 Deepwater Horizon (DWH) oil spill resulted in the oiling of approximately 2100 km of shoreline. During that time, resident organisms had to contend with the effects of multiple contaminants and suboptimal environmental conditions. This research examines the molecular effects of DWH oil spill contaminants in estuarine fish (CyprinodonvariegatusandFundulusgrandis) across multiple life stages, contaminant concentrations, and in conjunction with environmental stressors.

Our results indicate that:

1. Oil elicits substantial transcriptional effects across life stages in C. variegatus. In adults, exposure to low concentrations of oil and dispersant results in transcriptional effects related …

Investigating The Antiviral Effects Of Vig-3 From Rainbow Trout, Shanee Herrington-Krause Jan 2019

Investigating The Antiviral Effects Of Vig-3 From Rainbow Trout, Shanee Herrington-Krause

Theses and Dissertations (Comprehensive)

Rainbow trout is the most farmed fish in Ontario, and thus is economically important to the province. Despite this, there is a lack of understanding regarding fish innate immunity, specifically with regards to interferon-stimulated genes (ISGs) and their antiviral effector functions. ISGs are the workhorses of the innate antiviral response, operating together to limit each step of virus replication. The Viral Hemorrhagic Septicemia Virus (VHSV) induced gene (Vig)-3 is a newly identified ISG within many fish species and is homologous to ISG-15 in mammals. It is a small ubiquitin-like protein inducible by type I interferon (IFN-I), and is suggested to …

Nanoparticle Toxicity And Molecular Mechanisms In Fish: A Case Study With Silver Nanoparticles, Jiejun Gao Dec 2016

Nanoparticle Toxicity And Molecular Mechanisms In Fish: A Case Study With Silver Nanoparticles, Jiejun Gao

Open Access Dissertations

Nanoparticles (NPs) are widely used in a myriad of commercial and industrial products making their entry to the environment a likely event. NPs have unique physical-chemical properties that result from their small size and high surface area to volume ratio, making them highly reactive and potentially toxic. In Chapter 1, we summarize the effects and mechanisms of metal-based NPs on the vascular system. In vitro studies have shown that NPs are anti-angiogenic because they cause inflammation, oxidative stress, and apoptosis of endothelial cells resulting in increased permeability and decreased proliferation and migration. Whole animal studies examining the effects of NPs …
