Intrerconversion Of Replicative Forms Of Sindbis Virus Rna And The Electrophoresis Of Viral Rna, Jonathan D. Martin

Three species of double-strand (ds) RNA, designated RF I, RF II, and RF III in order of decreasing size, were produced by RNase treatment of extracts of chicken embryo cells infected for 6 h with Sindbis virus. These species were identifiable with those reported by Simmons and Strauss (28). At a low level of enzyme (0.001 µg/ml) the major species obtained was RF I. When the enzyme concentration was increased 10-, 100, and 1000-fold, there was a progressive increase in the proportions of RF's II and III and a concurrent decrease in the proportion of RF I. Cell-Mediated Cytotoxicity Of Jensen Tumor In Sprague-Dawley Rats, Theodore D. Masek

An in vitro system for the study of the cell-mediated immune response to Jensen Sarcoma (JS) is described. The tumor grew unrestricted and lead to the demise of 80% of successfully challenged Sprague-Dawley rats (persistors) while 20% of challenged rats (regressors) destroyed the tumor by an unknown mechanism(s). Lymphocytes from both persistors and regressors were shown to be cytotoxic for tumor cells in vitro. Pretreatment of tumor target cells with "immune" serum from persistors or regressors had no effect on the in vitro cytotoxicity of lymphocytes from tumor-challenged rats. Pretreatment of lymphocytes with irmnune serum, however, was shown to reduce …

The possible effects of ablastin on Schistosoma mansoni were investigated by (1) establishing a concurrent infection of Trypanosoma musculi and Schistosoma mansoni in mice, and (2) by obtaining Trypanosoma lewisi immune rat serum (ablastin) and injecting it into S. mansoni infected mice. These procedures were followed on days 4, 14 and 28 of S. mansoni infection. For the groups that were given immune rat serum, controls were kept into which normal rat serum was injected.

Ablastin does not have any effect on the development or morphology of the S. mansoni worms as tested in this study. Although ablastin inhibits glycolytic …

The assay of Bursa-derived and Thymus-derived lymphocytes (B and T cells) in man has become increasingly useful during the past few years in normal and in clinical conditions. There has been quite a range reported in the literature in the percent of B and T cells in normal subjects by different investigators.

The purpose of this study was to see if monocyte contamination was one cause of non-specific rosette formation. Dormant Tilletia caries teliospores in fixative solution or distilled water were frozen onto specimen chucks of an FTS Sorvall- Christensen frozen thin sectioner and cut or fractured at various temperatures (-20 to -75 C) and thickness settings (10, 15, 20, and 25 um). Cytoplasm of dormant spores was well preserved and organelles were found to differ from those of germinated spores in morphology. Irregular electron dense and electron opaque areas of lipid bodies were sometimes evident in dormant and germinating Tilletia caries teliospores. The Effect Of Urea On The Sedimentation Coefficient Of The Curly Top Virus Dimer, Allen H. Smith

The purpose of this project is to investigate the effect of increasing concentrations of urea on the sedimentation rate of the curly top virus dimer as measured by ultracentrifugation techniques. In general, urea causes the breaking of hydrogen bonds in macromolecules--in the case of the virus, increased concentrations of urea should cause changes in the configuration of the virus and may possibly cause a separation of the dimer into monomers. From previous centrifugation studies, we know that the dimer has a characteristic sedimentation coefficient(S) of about 80 Svedbergs; the monomer has an S value of about 55 Svedberg units. Increased …
