Background: Periodontal disease is one of the most prevalent oral health problems in Indonesia that affects supporting tissues of the teeth. Porphyromonas gingivalis plays an important role in the pathogenesis of periodontal disease. Alternative therapy with natural plant extracts, including pineapple (Ananas comosus) hump extract may inhibit the growth of bacteria that cause periodontal disease. Objective: To determine the effect of pineapple hump extract on bacterial growth and adhesion of Porphyromonas gingivalis biofilms. Background: Porphyromonas gingivalis adherence to Streptococcus gordonii may be important for P. gingivalis colonization in the oral cavity. Nanoparticles encapsulating synthetic peptide BAR (BAR-NPs) inhibit P. gingivalis adherence more potently than free BAR. However, BAR-NPs would exhibit low retention in an open flow environment. Hypothesis: Targeting BAR-NPs to the streptococcal surface using CafA protein will enhance their efficacy. Methods: CafA-modified NPs encapsulating BAR were synthesized using double emulsion approach. Surface binding and retention, and release kinetics of BAR from CafA-modified NPs was assessed. BACKGROUND: Periodontitis is an inflammatory disease caused by poly-microbial infection that leads to destruction of connective tissue and alveolar bone. It is well documented that bacteria-derived virulent factors that can act on Toll-like receptors (TLRs), represented by Lipopolysaccharides (LPS), are engaged in the initiation of inflammatory responses. A recent study demonstrated an association between the Porphyromonas gingivalis (Pg)infection and the progression of ESCC (Gao et al., 2016). However, how Pg infection affects the nature of ESCC remains unknown. We examined the effects of Pg on the proliferation and chemotherapy drug-induced apoptosis of ESCC cells and elucidated the signaling molecules involved in these processes. OBJECTIVE: The aim of this study was to compare the production of the chemokines CCL3 and CXCL12 by cultured dental pulp fibroblasts from permanent (PDPF) and deciduous (DDPF) teeth under stimulation by Porphyromonas gingivalis LPS (PgLPS). MATERIAL AND METHODS: Primary culture of fibroblasts from permanent (n=3) and deciduous (n=2) teeth were established using an explant technique. After the fourth passage, fibroblasts were stimulated by increasing concentrations of PgLPS (0-10 µg/mL) at 1, 6 and 24 h. MicroRNAs (miRNAs) are short, noncoding RNAs involved in the regulation of several processes associated with inflammatory diseases and infection. Bacterial infection modulates miRNA expression to subvert any innate immune response. In this study we analyzed, using microarray analysis, the bacterial modulation of miRNAs in bone marrow-derived macrophages (BMMs) in which activity was induced by infection with Porphyromonas gingivalis The expression of several miRNAs was modulated 3 h postinfection (at a multiplicity of infection of 25). Porphyromonas gingivalis, a Gram negative anaerobe implicated in the progression of periodontal disease, is capable of surviving and causing infection despite high levels of reactive nitrogen species found in the oral cavity due to its efficient nitrosative stress response. HcpR is an important sensor-regulator that plays a vital step in the initiation of the nitrosative stress response in many Gram negative anaerobic bacteria. Studies suggest that P. gingivalis functions as a keystone pathogen and interacts with primary colonizers in the supragingival biofilm such as S. gordonii. This interaction contributes to the initial colonization of the oral cavity by P. gingivalis and thus represents a potential target for therapeutic intervention. We have identified a peptide (BAR) derived from the streptococcal SspB protein that functions to inhibit P. gingivalis adherence to S. gordonii. Smoking is an established risk factor for periodontitis. Prior studies have shown that cigarette smoke extract (CSE) can induce profound phenotypic changes in Porphyromonas gingivalis and alters the virulence of this important periodontal pathogen. We hypothesized that CSE might also alter gene expression in established periodontal pathogens, Porphyromonas gingivalis and Treponema denticola, as well as in the emerging pathogen, Filifactor alocis. Although they reside in separate environments, they are both subject to hydrogen peroxide stress and have mechanisms to regulate the stress. OxyR is the primary transcriptional regulator/sensor of oxidative stress response caused by hydrogen peroxide. OxyR in P. gingivalis is not well-characterized compared to OxyR in E. coli. A gingival crevice model (epithelial cell-Porphyromonas gingivalis-neutrophil) was established and used to profile gingipain, matrix metalloproteinase (MMP), MMP mediators [neutrophil gelatinase-associated lipocalin (NGAL) and tissue inhibitor of metalloproteinases 1 (TIMP-1)] and cytokine networks. Smoking is the primary environmental risk factor for periodontitis. Therefore, the influence of cigarette smoke extract (CSE) was also monitored in the same model. Porphyromonas gingivalis alone induced low levels of interleukin-1b and interleukin-8 from epithelial cells, but high levels of both cytokines were produced on the addition of neutrophils. BACKGROUND: Considering that grafted gingival tissue might have to be adapted to the receptor area and that fibroblasts have the ability to respond to bacterial stimuli through the release of various cytokines, this study investigated whether fibroblasts from the palatal mucosa behave differently when grafted onto the gingival margin regarding cytokine secretion.

Gingipain activity in Porphyromonas gingivalis, the major etiological agent in adult periodontitis, is post-translationally regulated by unique Vim proteins including VimF, a putative glycosyltransferase. To ascertain the VimF mediated phenotype we first inactivated the vimF gene in P. gingivalis ATCC 33277 (FLL476), a less virulent fimbriated strain. We observed that the vimF-defective mutant (FLL476) showed a phenotype similar to that of the vimF-defective mutant (FLL95) in the P. gingivalis W83 background. While hemagglutination was not detected and autoaggregationn was reduced, biofilm formation was increased in FLL476. Also, invasive capacity decreased for this mutant. Oral bacterial biofilms trigger chronic inflammatory responses in the host that can result in the tissue destructive events of periodontitis. However, the characteristics of the capacity of specific host cell types to respond to these biofilms remain ill-defined. This report describes the use of a novel model of bacterial biofilms to stimulate oral epithelial cells and profile select cytokines and chemokines that contribute to the local inflammatory environment in the periodontium. Porphyromonas gingivalis is a major etiological agent that is responsible for the cause and progression of periodontal diseases. The bacterium is exposed to various environmental conditions and oxidative stress conditions while it is in the oral cavity. So, P. gingivalis should have an efficient regulatory system in order to adjust and survive in the oral cavity. Periodontal disease has been recently linked to a variety of systemic conditions such as diabetes, cardiovascular disease, preterm delivery, and oral cancer. The most common bacteria associated with periodontal disease, Porphyromonas gingivalis (P. gingivalis) has not yet been studied in the malignant gingival tissues. The objective of this study was to investigate the presence of P. gingivalis in specimens from squamous cell carcinoma patients. Matrix metalloproteinase-9 (MMP-9) cleaves collagen, allowing leukocytes to traffic toward the vasculature and the lymphatics. When MMP-9 is unregulated by tissue inhibitor of metalloproteinase-1 (TIMP-1), this can lead to tissue destruction. Dendritic cells (DCs) infiltrate the oral mucosa increasingly in chronic periodontitis, characterized by infection with several pathogens including Porphyromonas gingivalis. In this study, human monocyte-derived DCs were pulsed with different doses of lipopolysaccharide of P. gingivalis 381 and of Escherichia coli type strain 25922, as well as whole live isogenic fimbriae-deficient mutant strains of P. gingivalis 381. Purpose: The biomedical application of low temperature atmospheric pressure plasma (LTAPP) is a collaborative interest for engineering, medical, dental and biological researchers. This laboratory study tested whether LTAPP can limit growth of Porphyromonas gingivalis, a periodontal pathogen strongly associated with periodontal disease, disease progression, and refractory periodontitis. Methods: After extensive pilot trials, the study consisted of 24 agar plate samples of P. gingivalis- 20 samples were exposed to LTAPP at 5, 7, 9 and 10 minutes and 4 control samples were exposed to helium gas only. The aim of the present investigation is to purify, characterize, and determine the chemical and biological properties of Porphyromonas gingivalis free dihydroceramides (fDHC). Effects were tested by examination of human gingival fibroblast morphologic changes in addition to their synthesis of prostaglandin E2 (PGE2) and prostaglandin F2α (PGF2&#;) produced in vitro. Free dihydroceramides as well as phosphoglycerol dihydroceramide (PGDHC), and de-esterified phosphoglycerol dihydroceramide (PGDHC-C15:0) were obtained by total lipid extraction of P. gingivalis followed by high performance liquid chromatography (HPLC). The efficacy of a novel synthetic antimicrobial peptide (WLBU2) was evaluated against three oral microorganisms (grown planktonically): Streptococcus gordonii, Fusobacterium nucleatum, and Porphyromonas gingivalis. WLBU2 killed all three species, with F. nucleatum being the most susceptible. WLBU2 also reduced the bacterial burden of S. gordonii and F. nucleatum biofilms.

การระบุเชื้อพอร์ฟีโรโมแนส จิงจิวาลิส ที่นํามาจากร่องลึกปริทันต์อักเสบชนิดแอดวานซ์ โดยวิธีพีซีอาร์, จินตกร คูวัฒนสุชาติ

วัตถุประสงค์ การศึกษาครั้งนี้ ต้องการนําวิธีพีซีอาร์ มาใช้ยืนยันการระบุเชื้อพอร์ฟีโรโมแนส จึงจิวาลิส ที่นํามาจาก ร่องลึกปริทันต์อักเสบชนิดแอดวานซ์ของผู้ป่วยจํานวน 3 ราย วัสดุและวิธีการ ทําการแยกเชื้อแบล็คพิคเม็นเต็ด แบคทีรอยดีส (3 ชนิด ประกอบด้วยเชื้อหมายเลข 1, 2 และ 3) จากเชื้อที่ได้จากร่องลึกปริทันต์ของผู้ป่วยโรคปริทันต์อักเสบชนิดแอดวานซ์ 3 ราย โดยผู้ป่วยเป็นชาวไทยที่มารับ การรักษาโรคปริทันต์จากภาควิชาปริทันตวิทยา คณะทันตแพทยศาสตร์ จุฬาลงกรณ์มหาวิทยาลัย ทําการเพาะเลี้ยง เชื้อที่แยกมาได้ (ทั้ง 3 ชนิดนี้) ลงบนจานอาหารเลี้ยงเชื้อพวกเบรนฮาร์ทอินฟิวชั่นยีสต์เอ็กแทรกบลัดชีพอาการใน ตู้อบที่ปราศจากออกซิเจนที่มีไนโตรเจนร้อยละ 80 ไฮโดรเจนร้อยละ 10 และคาร์บอนไดออกไซด์ร้อยละ 10 ที่อุณหภูมิ 37 องศาเซลเซียส เป็นเวลา 10 วัน แล้วนําเชื้อแบล็คพิคเม็นเต็ด แบคทีรอยดีส (ทั้ง 3 ชนิดที่เพาะเลี้ยง ขึ้นเรียบร้อยแล้ว) ไปทําการแยกดีเอ็นเอ สําหรับการทําพีซีอาร์ เพื่อทําการระบุชนิดของเชื้อต่อไป ผลการศึกษา ผลที่ได้จากการศึกษา ปรากฏว่า เชื้อแบล็คพิคเซ็นเต็ด แบคทีรอยดีส 3 ชนิด ที่ประกอบด้วยเชื้อ หมายเลข 1, 2 และ 3 นั้น เชื้อหมายเลข 1 กับเชื้อหมายเลข 2 ให้แถบกับไพรเมอร์พีจี (19+20) กับไพรเมอร์พีจี (21+22) ซึ่งตรงกับกลุ่มควบคุมบวกที่ให้แถบกับไพรเมอร์พีจี (19+20) กับไพรเมอร์พีจี (21+22) ส่วนเชื้อหมายเลข 3 ไม่ให้แถบกับไพรเมอร์พีจี (19+20) กับไพรเมอร์พีจี (21+22) สรุป วิธีพีซีอาร์เป็นวิธีที่เหมาะต่อการนํามาใช้ระบุเชื้อพีจีจากผู้ป่วยด้วยโรคปริทันต์อักเสบ เพราะสามารถระบุเชื้อพีจีได้รวดเร็วและแม่นยํา (ว ทันต จุฬาฯ 2549;29:161-170)
