In female sheep, the release of gonadotropin-releasing hormone (GnRH) from the hypothalamus triggers luteinizing hormone (LH) to be released from the pituitary. Luteinizing hormone, in turn, acts on the ovaries to elicit estradiol (E2) release. E2 acts in a negative feedback manner to control the amount of LH released. The increase in LH release that drives puberty onset results from a decrease in sensitivity to E2 as the animal ages. However, the neural mechanisms mediating this change in sensitivity are not known, as is where in the hypothalamus E2-negative feedback occurs. In the arcuate …

Nicotine dependence is causally linked to increased risk of mood/anxiety disorders in later life. Females are reported to experience a higher prevalence of anxiety/depressive disorders and challenges in smoking cessation therapies, suggesting a potential sex-specific response to nicotine exposure and mood/anxiety disorder risk. However, pre-clinical evidence of sex-specific responses to adolescent nicotine exposure is unclear. Thus, to determine any sex differences in anxiety/depressive-related outcomes, adolescent male and female Sprague Dawley rats received nicotine (0.4 mg/kg; 3x daily) or saline injections for 10 consecutive days, followed by behavioural testing, in-vivo electrophysiology and Western Blot analyses. Resting cerebral blood flow (CBF) is increased during the luteal (LUT) phase of the menstrual cycle; however, it is unclear whether this change impacts a postexercise executive function (EF) benefit. Female participants (N=16) performed three experimental sessions: a V̇O2peak task and 20-min single bouts of moderate intensity aerobic exercise (i.e., 80% of lactate threshold) during their follicular (FOL) and luteal (LUT) menstrual cycle phases. A separate group of male participants (N=21) additionally completed a V̇O2peak test and a 20-min exercise intervention. Non-small cell lung cancer (NSCLC) remains the leading cause of cancer-related deaths worldwide. Targeted therapeutic agents, such as epidermal-like growth factor receptor (EGFR) tyrosine kinase inhibitors (TKIs) or monoclonal antibodies targeting vascular endothelial growth factor (VEGF/R), can effectively inhibit upregulated signaling pathways driving tumorigenesis in NSCLC and many other cancers. Unfortunately, however, resistance to such targeted therapies inevitably arise in most patients and can occur through a variety of resistance mechanisms including genomic alterations and upregulation of bypass pathways. Circadian rhythms are approximately 24-hour cycles of behavior, physiology, and gene expression. In mammals, these circadian rhythms are generated by clocks located in nearly every tissue in the body. The function of circadian clocks is to synchronize physiology and behavior with environmental cycles such as the light-dark cycle. After menopause, when circulating levels of estrogens are very low, women are more susceptible to obesity comorbidities such as metabolic syndrome and cardiovascular disease, suggesting that estrogens regulate these processes. Estrogens could protect females from metabolic dysfunction by regulating circadian rhythms. Consumption of diets that are high in fat contribute to obesity. …

Women are far more likely to develop anxiety and depression than men. It is believed that the dysregulation of the HPA axis by the binding of corticotropin -releasing factor (CRF) to the corticotropin-releasing factor receptor 1 (CRFR1) contributes to the likelihood of these stress- related disorders. Estrogens acting through Estrogen receptor alpha (ERa) have been shown to increase anxiety production upon activating the HPA axis. Chlamydia is the most common bacterial sexually transmitted infection in the United States and worldwide. It often goes unnoticed due to lack of symptoms and left untreated it can ascend the female genital tract to cause sequelae like pelvic inflammatory disease and irreversible tubal infertility. This behavioral diversity is due to the complex integration of external and internal stimuli, through both neuronal and hormonal signals that selects the best behavioral response. Pharmaceuticals, including hormones and antibiotics, are considered contaminants due to their widespread use and release into the environment. Hormones, like the synthetic estrogen used in oral contraceptives (17α-ethinylestradiol), are present in freshwater and marine systems, but with relatively unknown effects on the organisms that live there. Ethinylestradiol (EE2) accumulates in waterlogged soil (sediment) with potential to harm sediment-dwelling animals. For example, fiddler crabs (Uca pugilator) are vital members of salt marsh communities. Cadmium, a heavy metal and carcinogen, is an environmental and workplace contaminate. As a known endocrine disruptor, it can mimic the proliferative effects of estrogen and is classified as a metalloestrogen. While the proliferative effect of estrogen on cancerous cell growth has been well established, the effects of cadmium have not been fully examined. To determine if cadmium stimulates growth in two human ovarian adenocarcinoma cell lines, OVCAR3 and SKOV3, cells were treated for 48 hours with varying concentrations of cadmium, 0.001 µM – 10 µM, and growth was measured using a cell proliferation assay. The purpose of this study was to examine sex-specific differences in the blood glucose (BG) response to recurrent aerobic exercise in type 1 diabetes. Specifically, we examined the role of peak estrogen (E2) concentrations during proestrus on BG response to prolonged aerobic exercise. To do so, nineteen Sprague-Dawley rats were assigned to four exercised groups: control female (CXF; n = 5), control male (CXM; n = 5), diabetic female (DXF, n = 5) and diabetic male (DXM, n = 4). The hypothalamic-pituitary-gonadal (HPG) axis is responsible for the production of the hormones testosterone and estradiol, and testosterone is thought to contribute to regulation of the axis through a negative feedback mechanism. Regulation by negative feedback involves the product of a pathway turning off that pathway when enough product is made. However, because the enzyme P450 aromatase converts testosterone to estradiol, estradiol may also contribute to regulation of the HPG axis and other phenomena that have been attributed to testosterone, like the inhibition of immune function. Addiction is characterized by a progressive loss of executive control over drug-seeking and consumption, and may be associated with a behavioral shift from instrumental goal-directed actions to stimulus-response habits. Sex differences in drug addiction have been linked to changing hormone levels across the estrous cycle, and females exhibit a particular vulnerability to psychostimulants such as cocaine and amphetamines. Bisphenol A (BPA) is a well-known endocrine disrupting chemical that mimics the effects of estrogens. Aromatase B (Cyp19a1b) is a brain-specific enzyme that converts testosterone to estrogen and is highly upregulated in response to estrogen receptor activation localized to radial glial cells. During embryonic zebrafish development, there is a small window of time denoted by an increase in neurogenesis and estrogen receptor activity. Although several sex differences have been described in brain structure, function, and development, sex as a biological factor is underrepresented in neuroscience studies. In the mammalian brain, there are sex differences in the mechanism of rapid estradiol actions on neuronal physiology. In the songbird, the brain is a major source of estradiol production, and estradiol rapidly modulates auditory responsiveness through dynamic changes and an unknown receptor mechanism. Estradiol (E2) has been found to influence dopamine (DA) activity in the nonhuman animal brain. While there has been very little research performed looking at E2’s effects on DA-related cognitive function in humans, recent research found that women tested during high E2 phases of the menstrual cycle had significantly better performance on a DA-dependent spatial working memory task, than women tested during the lowest E2 phase. PURPOSE: There is evidence that female sex hormones impact caffeine metabolism, with decreased CYP1A2 activity and diminished caffeine clearance in women with higher estrogen levels. Therefore, the objectives of this project were to determine the effects of oral contraceptives and menstrual cycle on the benefits of caffeine supplementation for cycling performance. The current study explored the possibility that maternal estrogen dominance serves as a risk factor for having a child with autism. An online survey was sent to both biological mothers of autistic children (n=253) and biological mothers of non-autistic children (n=221). The survey presented a series of questions pertaining to both endogenous and exogenous factors and exposures that could increase estrogen levels. Despite converging incidence rates for breast cancers by race, disparities in mortality persist where black women suffer from poorer prognosis compared to white counterparts. To understand the clinical, demographic, and molecular characteristics underlying these disparities, we examined differences among patients with breast cancer to understand the role of human epidermal growth factor receptor 2 (HER2) status, age, and race/ethnicity among women diagnosed with hormone receptor-positive breast cancer, and disparities in surgical therapy among female patients with early stage young-onset breast cancer. Estradiol, traditionally known as a hormone that communicates with distant cells in the body, is also synthesized locally in the brain to act as a neuromodulator. Neuromodulators differ from neurotransmitters in that they simultaneously affect a population of neurons and their actions are not limited to the synapse. One of the many effects of estradiol signaling is rapid modulation of auditory processing in response to external stimuli. Nanomolar concentrations of both cadmium and arsenite, two environmental estrogens present in cigarette smoke, have been documented in rapidly phosphorylating ERK1/2, a type of MAPK, in the human non-small cell lung cancer (NSCLC) line NHI-1793 in a manner similar to that of estrogen. Pretreatment of cells with a general, nonspecific estrogen receptor antagonist reduced the levels of phosphorylated MAPK, indicating that this phosphorylation event is achieved through use of an estrogen signaling pathway. Estrogen is involved in the proliferation of adenocarcinoma lung cells. However, the proliferative role of two endocrine disrupters, cadmium chloride and sodium arsenite, in lung adenocarcinoma is less clear. Previous studies have shown that these two heavy metals can induce cellular proliferation and activate phosphorylation of ERK1/2 in a human lug adenocarcinoma cell line, NCI-H1793. Induced phosphorylation of ERK1/2 by these compounds is decreased in the presence of an estrogen receptor antagonist, ICI-182,780, suggesting that cadmium and arsenite stimulate these responses, in part, via a non-genomic estrogen signaling pathway. Estrogens are a class of hormones that are demonstrated to be neuroprotective. The levels of estrogen in the body decline during menopause which can cause a variety of symptoms. Hormone replacement therapy (HRT) is a treatment option for menopause, however, it has been demonstrated to have adverse side effects. No direct, quantitative, and non-invasive markers presently exist to assess exercise-induced muscle damage (EIMD). However, magnetization transfer ratios (MTR), an emerging measurement quantified via magnetic resonance imagery, provide more direct indication of muscle integrity following EIMD. This study compares and correlates the temporal pattern of the MTR to alternative indirect markers of EIMD in male vs. female populations, and subsequently establishes whether there are sex differences in biochemical activity during recovery. The antioxidant properties of estrogen hypothetically minimize muscle trauma, maintain membrane stability, and limit swelling resulting in heightened tissue integrity and resilience to EIMD. Thousands of synthetic chemicals have been released into the environment, causing widespread exposures of wildlife and humans alike. Some of these chemicals are known to disrupt aspects of hormone action thus inducing abnormalities in endocrine tissues and organs. Bisphenol S (BPS) and tetrabromobisphenol A (TBBPA), two largely unstudied chemicals commonly used in consumer products, are suspected to have endocrine disrupting properties based on their similar chemical structures to known endocrine disrupting chemicals (EDCs). To determine whether developmental exposure to BPS or TBBPA induced abnormalities in the ovary, mice were administered oil, BPS or TBBPA during pregnancy and lactation. The ovaries …
