Glutamate synthases (GOGAT) were analyzed to identify the functional binding domains of the substrate (glutamine) and cofactors (FMN, NAD(P)H, FAD, [3Fe-4S]1+,0 and [4Fe-4S]2+,1+ clusters and ferredoxin) on this enzyme. The published amino acid sequences of six different NAD(P)H-dependent GOGATs (NAD(P)H-GOGAT) and ten different ferredoxin-dependent GOGATs (Fd-GOGAT) were used for this analysis. The amino acid sequences of these sixteen GOGATs were compared with the amino acid sequences of aminotransferases for glutamine, flavoproteins for FMN, flavoproteins and pyridine-nucleotide-dependent enzymes for FAD and NAD(P)H, iron-sulfur proteins for [3Fe-4S] 1+,0 and [4Fe-4S]2+,1+ clusters and ferredoxin-dependent enzymes for ferredoxin. Production Of Uricase Enzyme From Aspergillus Niger And Determination Of Some Factors Affecting The Activity, Fi̇gen Ertan, Erol Aksöz

This study was undertaken to obtain uricase enzyme from Aspergillus niger and also to determine the production conditions and the effects of some factors on enzyme activity. The results of the experiments, in consideration of the production of the uricase enzyme and the factors affecting the production, demonstrated that enzyme activity was maximum when the production time was 3 days at a temperature of 30ºC with the initial pH at 6.0 and inductor concentration at 0.1%. Characterization Of \Beta-Xylosidase And\Alpha-L-Arabinofuranosidase Activities Fromthermomonospora Fusca Bd25, Müni̇r Tuncer

Thermomonospora fuscaBD25 produces relatively high levels of activity of \beta-xylosidase and \alpha-L-arabinofuranosidase. The aim of the work described in this study was to characterize some properties of \beta-xylosidase and \alpha-L-arabinofuranosidase produced by T. fusca BD25 when growing on oat spelt xylan as the main carbon and energy sources. The substrates p-NPX and p-NPA reacted with \beta-xylosidase and \alpha-L-arabinofuranosidase with specific activities of 4.01 U mg -1 protein and 0.35 U mg -1 protein, respectively. Apoptosis induced by hydrogen peroxide (H_2O_2 ) was investigated in Chinese hamster ovarian (CHO) cells, and compared to apoptosis induced by cisplatin (25 µM), a well-established apoptosis-inducing agent. In preliminary experiments, CHO cells were treated with low concentrations of H_2O_2 (250-500 µM) continuously or for 3 hours, followed by washing and further incubation in complete growth medium for 1-10 days. Fluorescence microscopy of acridine orange-stained cells indicated morphological changes of apoptosis. Similar changes were observed in cisplatin-treated cells. In this study, 18 live specimens collected from six localities in the distribution areas of Mesocricetus brandti and Mesocricetus auratus were examined using SDS-PAGE (sodium dodecyl sulphate-polyacrylamide gel electrophoresis). The findings showed that there is no difference in globulin and albumin proteins of M. brandti and M. auratus, and supported the claim that M. auratus is very closely related to M. brandti.

Sulphur dioxide as a criterion of air pollution supresses macrophage functions. Some immunological changes in diabetes mellitus have been detected. In the present study, we examined the effect of sulphur dioxide on macrophage phagocytic activity in diabetic rats. The animals were separated into four groups as: 1) Control, 2) Sulphur dioxide, 3) Diabetes mellitus, 4) Diabetes mellitus+sulphur dioxide. In this study, the population structure and growth properties of Chalcalburnus mossulensis (Heckel, 1843) caught from Askale Region of River Karasu betwen February 1996 and January 1997 were examined. The age composition of population varied between I and VI ages, and the most abundant age group was the third age group (42.13%). The population was composed of 51.74% females and 48.26% males. The fork length and the body weights of males and females varied between 8.9-18.1 cm, 3.6-67.9 g and 8.5-18.5 cm, 5.1-79.5 g respectively. The effect of some nutrients added to synthetic compost with wheat straw, on mycelium growth, yield and early ripe of Agaricus bitorquis (Quél.) Sacc. were investigated. The compost was prepared by fermentation and chemical disinfection methods. Corn flour, wheat flour, hen grain, soybean meal and sunflower seed hulls were used as additional nutrients. The experiments were made at the temperatüre 20-22°C and 80-90% relative humidity in the yield rooms. The purpose of this study was to determine the effects of gibberellic acid (GA 3 ) on male and female sterility, seed yield, oil and fatty acid syntheses, growth and development properties in safflower ( Carthamus tinctorius var. Yenice). GA 3 was applied in five different doses at three different stages, and induced male sterility at a rate of up to 93.0%. This finding has important implications for the practical use of GA 3 in hybrid seed production. In order to obtain high amounts of \alpha-amylase enzyme from Bacillus bacteria a new medium was developed. In this medium, three strains of Bacillus bacteria were grown in the presence of different carbon, nitrogen and metal ions. The highest \alpha-amylase yield was obtained by addition of Na-citrate, strach and sucrose for the strains of B. subtilis, B. amyloliquefaciens I and B. amyloliquefaciens II, respectively. The data compiled during five-year-long investigation on the influence of drilling water and crude oil on specific composition, number and structure of the communities of free-living soil ciliates is presented. The vertical distribution of ciliates and their colonizing potential in the sterile soil monoliths placed in clean and polluted soils was studied on the basis of their survival and physiological responses. The changes in certain ciliate species were demonstrated by biotesting of the influence of various concentrations of drillinig water, crude oil and drillinig muds on these ciliates.

Isolation, Characterization And Identification Ofthiram-Degrading Microorganisms From Soilenrichment Cultures, Nuretti̇n Şahi̇n, Abdurrahman Üsame Tamer

Mixed microbial cultures were obtained by enrichment from soil samples collected from the Gediz basin. A number of species of bacteria and fungi were isolated and partially characterized from enrichment clutures containing the fungicide thiram. According to their responses in morphological and biochemical tests fungi were identified as Aspergillus niger, A. flavus and Penicilium steckii. Bacterial isolates were assigned to the genera: Bacillus, Arthrobacter, Moraxella-like Acinetobacter and Streptomyces. Islates grew in a mineral salts medium containing 0.2 mg/ml thiram and no growth factors were required.

Olive oil mill effluent (OOME) is one of the major pollutants produced by the agricultural industry in Mediterranean countries. The olive oil mills in these countries generate about 2-3 million tonnes of effluent annually. OOME is a black liquor containing a water-soluble fraction of ripe olives and water that is used in the process of olive oil extraction. We report here the development of hybrid cells producing monoclonal antibodies specific for two different steroid hormones with mixed immunization using hybridoma technology. BALB/c mice were immunized with a mixture of three steroid antigens: progesterone, estradiole and testosterone linked to bovine serum albumine. These mice were used for fusion. In the two fusion experiments, ELISA tests showed that among 645 wells only 2 hybrids reacted with progesterone (MAM 3C2, MAM 3E3) and one of them reacted specifically with estradiol (MAM 7H7). Native-polyacrylamide gel electrophoresis (N-PAGE) was used to characterize Candida albicansstrains isolated from vaginal specimens of patients suffering from vaginitis in Ankara hospitals, Turkey. Three different protein band profiles were observed among these 38 C. albicansisolates when whole cell protein extracts of these strains were used in N-PAGE. 34 of the 38 isolates were shown to belong to a single group or clone with a dissemination percentage of 89, while three isolates belonged to another group with a percentage of 8. In this study, over-produce esterase allele types dependent on organophosphate resistance in mosquitoes were determined by cellulose acetate gel electrophoresis in the human malaria vector species Anopheles sacharovi and Culex pipiens in Çukurova (Adana). We found A_2 and A_5 allele bands with some unknown and unidentified gel bands in some An. sacharovi field samples, of which some bands seemed to match with over-produce esterase. We identified A_2-B_2 allele types for all the Cx. pipiens samples collected in this study.

In this study, microsomal nitrosodimethylamine demethylase (NDMAd) and cytoplasmic glutathione-s-tarnsferase (GST) activities in the livers of chicks obtained from the LD 10 (3 mg/ml) and LD 30 (9 mg/ml) doses of daminozide (Succinic acid 2,2-Dimethylhydrazide) injected Ross-Pm-3 fertilized chicken eggs were investigated. Plasmids encoding lactic acid fermentation ability were investigated in 50 strains of L. lactis subsp. lactis. Acriflavine was used for the elimination of lactose fermentation ability (Lac + ) from wild type strains. Lactose negative (Lac - ) mutants were selected on lactose indicator agar media. Six different lactose plasmids were identified for fifty strains by comparison of plasmid profiles of Lac - and Lac + mutants. The molecular sizes of these lactose plasmids ranged between 26.3 and 38.5 kb.

Some kinetic properties of hepatic lipase of Oncorhynchus mykisswere investigated. Hepatic triacylglycerol lipase was found to be located in the cytosolic fraction of the cell. Optimal activity of hepatic lipase occurred at pH 8.00 and 32 °C. When pNPA was used as a substrate, Vmax and Km values of hepatic lipase were estimated as 0.40 U/mg protein and 0.12 mM pNPA respectively. A stimulating effect of monovalent (NaCl and KCl) and divalent (CaCl 2 and MgCl 2 ) metal ions on hepatic lipase activity was determined. The effects of cadmium nitrate on the fecundity, larval survival and sex ratio of D. melanogaster were tested. According to our findings, the effects of cadmium nitrate can be summarized as follows: it appeared that when used as a feeding substance during the developmental period, cadmium nitrate did not have any effect on fecundity. On the other hand, if cadmium nitrate was given throughout adult life it caused a proportional decrease in the daily mean with increasing concentration. The cultural characteristics of Morchella esculenta mycelium were investigated in potato dextrose agar, malt extract agar and complete medium yeast plates prepared with casein (cas), Casamino Acids (caa), peptone (p) and sodium nitrate (NaNO 3 ). Aerial and vegetative mycelium types were determined in the agar media with cas, caa and p. The third colonial characteristic was observed in the agar media with NaNO 3 . In this media, a colonization increase in circular forms spreading from the centre to the edges was determined. Mycelium colonization periods were 20-24 days in agar media with NaNO 3 .

Nonspecific esterases in the muscle, heart, kidney and liver tissues of wild rats (Rattus norvegicus) were examined by horizontal starch gel electrophoresis. Zymogram bands corresponding to Es-10 and Es-4 in the muscle, heart, kidney and liver homogenates, Es-3 and Es-17 in the kidney and liver, Es-18 in the liver tissue, Es-15 in kidney and liver, Es-2 in the muscle and heart, Es-16 in the heart tissue were detected. These findings showed that nonspecific esterases are very polymorphic and tissue-specific, and are not of taxonomic importance for the populations of Rattus norvegicus.

The effects of N-nitrosopyrrolidine (NPYR) on mice chromosomes and micronucleus induction were investigated in order to evaluate its genotoxic effects. Twenty 10-12 week old mice and 40 8- 10 week old male mice ( Mus musculus var. albinos) were used for chromosome analysis without sex discrimination and micronucleus tests respectively. Mice were intraperitoneally treated in single injections with three dose levels (100, 200 and 300 mg/kg) of NPYR with isotonic saline as the solvent vehicle. In this study, the composition and density of the benthic algae and seasonal variations of the benthic algae of Palandöken Pond were determined between September 1990 and September 1992. Samples were collected from two previously selected sampling sites. It was found that Bacillariophyta was the dominant benthic algal group in this pond. During our investigation a total of 160 taxa were identified and 135 them were composed of epipelic algae. The percentages of algal groups were as follows: Bacillariophyta 57%, Cholorphyta 11% and Euglenophyta5%. In this study, the monthly variation of blood glucose level of Capoeta tinca caught from in Çoruh Basin - Oltu Stream, between Augist 1995 and June 1996 was determined. It was determined that the average blood glucose level was 86.19±3.86 mg/dl in males and 83.58±3.77 mg/dl in females. The blood glucose level was highest (116.36±8.58 mg/dl) in April, and lowest (51.00±7.10 mg/dl) in June.
