Verdicchio is a white grape varietal notably grown in the Marche wine region of east-central Italy within two Denominazione Origine Controllata (DOCs); Castelli di Jesi and Matelica, located less than 50 km apart. Jesi lies in the Apennine foothills near the coast of the Adriatic Sea and produces a smooth wine noted with apple-pear undertones and crisp, clean acidity. Conversely, Matelica is located in a synclinal valley of a fold-and-thrust belt and produces stronger wine with penetrating floral aromas and bold acidity. While macro-climatic variations do exist between DOCs, this research focuses on geological variations, specifically variations in soil composition, …

Over 10,000 Americans are killed in firearm homicides each year, and an additional 40,000 are injured in nonfatal shootings. There is a significant public health need to identify risk factors that can be modified to prevent firearm violence. Environmental lead exposure is a demonstrated neurotoxicant which causes behavior changes that are known to be criminogenic. More recent research has demonstrated that homicides and nonfatal shootings differ by the circumstances that lead to the shootings (i.e. gang, domestic violence, arguments) and aggregating them could lead to biased results. Common oat (Avena) has held a significant place within the global crop community for centuries; although its cultivation has decreased over the past century, its nutritional benefits have recently garnered increased interest for human consumption. No published reference sequences are available for any of the three oat subgenomes. Here we report a quality sequence assembly, annotation and hybrid optical map of the A-genome diploid Avena atlantica Baum and Fedak. The assembly is composed of a total of 3,417 contigs with an N50 of 11.86 Mb and an estimated completeness of 97.6%. This genome sequence will be a valuable research tool …

Human-bear interactions are an important consideration of bear biology, as interactions can lead to destruction of property as well as injury or death for both human and bear. Successful analysis of why these interactions occur can lead to appropriate preventative measures and mitigation of further conflict. Bryce Canyon National Park (BRCA) is comprised of relatively poor bear habitat, but a black bear population exists on the Paunsaugunt Plateau, on which the park occupies the eastern edge. Introduction: Irish dance has evolved to become more athletically demanding, thus making the art form very hard on dancers' bodies. Irish dancers must land from difficult moves without letting their knees bend or heels touch the ground, causing large amounts of force to be absorbed by the body. Past studies have found dancers landing with a range of 4.5–6 times body weight, potentially causing high amounts of overuse injury. The average lifespan is constantly increasing with the advent of new medical techniques, and age-related cognitive decline is becoming a prevalent societal issue. Even during healthy aging, humans and rats exhibit progressive deficits in episodic/declarative memory. In laboratory rats, age-related memory impairment can be assessed with trace fear conditioning (TFC). Recent research implicates ubiquitin proteasome system-mediated protein degradation in the synaptic plasticity supporting memory formation and retrieval. An important obstacle to a functional cure for HIV/AIDS is the persistence of viral reservoirs found throughout the body in various cells and tissues. Reservoirs can be latently infected cells, or in the case of follicular dendritic cells (FDC), non-infected cells that trap infectious virus on their surface through immune complexes (HIV-IC). Although several strategies have been employed to target and eliminate viral reservoirs, they are short-lived and ineffective. Dendritic spine plasticity is thought to be essential for the formation and storage of memories. The sex-steroid hormone 17-estradiol (E2) increases dendritic spine density in 2 brain regions necessary for memory formation, the dorsal hippocampus (DH) and medial prefrontal cortex (mPFC), but the mechanisms through which it does so remain largely unknown. Further, the extent to which these brain regions interact to mediate E2’s effects on memory is also unclear. The first part of my thesis is focused on a new family of two-component response regulator proteins: Aspartate-Less Regulators (ALR). They lack the catalytic aspartate residue required for the phosphorylation mechanism of typical two component response regulators. We are using biophysical tools to characterize two proteins with redox-sensitive ALR domains: repressor of iron transport regulator (RitR) from Streptococcus pneumonia R6 and diguanylate cyclase Q15Z91 from Pseudoalteromonas atalantica. For millions of years, cold water temperatures have excluded shell-crushing (durophagous) predators from continental-shelf environments off the western Antarctic Peninsula. Recently, however, king crabs, Paralomis birsteini, have been found in dense, reproductively viable populations on the upper continental slope, and rapid warming might enable them to expand to the shelf. King crabs are crushing shelled invertebrates on the slope, but calcification of sturdy, shell-crushing chelae is likely inhibited by the low saturation state of high-Mg calcite in the deep Southern Ocean. Phosphate is an essential compound for life. Escherichia coli employs a signal transduction pathway that controls the expression of genes that are required for the high-affinity acquisition of phosphate and the utilization of alternate sources of phosphorous. These genes are only expressed when environmental phosphate is limiting. The seven genes for this signaling pathway encode the two-component regulatory proteins PhoB and PhoR, as well as the high-affinity phosphate transporter PstSCAB and an auxiliary protein called PhoU. As the sensor kinase PhoR has no periplasmic sensory domain, the mechanism by which these cells sense environmental phosphate is not known. This paper …

PURPOSE: The aim of this study was to investigate the acute effects of various intensities of volume matched resistance exercise with blood flow restriction (BFR) and traditional resistance exercise on hemodynamic variables, arterial compliance, oxygen consumption (VO2), carbon dioxide production (VCO2), respiratory exchange ratio (RER), and energy expenditure (EE) during exercise and after exercise.

RESULTS: Significant condition main effects (p < 0.05) were found for RPE, HR, crPWV, fdPWV, LAE, SAE, TVI, RER during and postexercise. BFR25 cause greater increases in RPE and HR. BFR50 caused marked increases in SAE, crPWV, and fdPWV.

CONCLUSION: All BFR and traditional exercise conditions found no significant differences in EE, VCO2, and VO2. The lack of differences is likely due to time of protocol. Further protocols should implement greater time of exercise, as BFR had increased hemodynamic …

The Texas Parks and Wildlife Department Artificial Reef Program has deployed over 2,800 hectares of reefing area composed of petroleum platforms, steel vessels, and concrete. Artificial reefs have been shown to support many important reef fishes such as red snapper (Lutjanus campechanus), however, few studies have compared fish behavior on artificial reefs and natural habitats. We examined activity levels, behavioral patterns and habitat preferences of red snapper over natural reefs, oil platforms and submerged ships. In 2010, researchers observed an abrupt decline in nesting numbers of the Kemp’s ridley following 35 years of positive growth. In wake of this halt, and subsequent lowered nesting averages, evaluation of the genetic diversity of remaining individuals is relevant to investigate the plausibility of a genetic bottleneck, and determine the reproductive stability of the Kemp’s ridley.

The calanoid copepod, Acartia tonsa is one of the most abundant and well-studied estuarian species. However, the idea that this cosmopolitan species has unrestricted dispersal and high gene flow has been challenged. In this study, a more holistic picture of the phylogeography of A. tonsa was developed using the mitochondrial gene cytochrome oxidase one (mtCOI). Multiple new lineages were found in the Texas Gulf of Mexico that are basal to Northeastern Atlantic lineages. Connectivity was also observed between Brazil and the Texas Gulf of Mexico. The PI3K pathway controls many cellular processes such as cell growth, survival, metabolism, apoptosis, and differentiation. Canonically, PI3K activates AKT leading to subsequent nuclear exportation of FOXO transcription factors. Dysregulation of the PI3K pathway is a hallmark in many cancers and often is attributable to a null mutation in PTEN. PTEN is a phosphatase that reverses the reactions catalyzed by PI3K. Symptomology includes dermatological manifestations such as discoid lesions, acute cutaneous rashes, and oral and nasal ulcers, along with musculoskeletal, pulmonary, and renal complications. Abnormal T and B lymphocyte function and apoptosis, immune complex clearance, complement function, and nucleosome processing are typical of disease pathophysiology. Research in our lab has predicted hundreds of bacterial genes that influence nine different traits in the fruit fly, Drosophila melanogaster. As a practical alternative to creating site-directed mutants for each of the predicted genes, we created an arrayed transposon insertion library using a strain of Acetobacter fabarum DsW_054 isolated from fruit flies. Creation of the Acetobacter fabarum DsW_054 gene knock-out library was done through random transposon insertion, combinatorial mapping and Illumina sequencing. Successful mapping of transposon insertion was achieved for 6418 mutants with hits within 63% of annotated genes within Acetobacter fabarum DsW_054. Insertion sites were verified in …

Current literature on landing biomechanics in a CAI population, however, considers all patients as a homogeneous group. Synapses can be altered due to experiences in a process called synaptic plasticity, which causes memory formations. Synapses can be strengthened through methods known as long-term potentiation (LTP) or weakened through long-term depression (LTD). Stresses can cause changes by altering synapses through either LTP or LTD. Rats were used to study the effects of post-traumatic stress disorder (PTSD)-like symptoms and a prophylactic treatment using pharmaceuticals. The first model used was the single prolonged stress (SPS) with two weeks of chronic light, which was not as effective for causing changes in synaptic plasticity. Synapses can be altered due to experiences in a process called synaptic plasticity, which causes memory formations. Synapses can be strengthened through methods known as long-term potentiation (LTP) or weakened through long-term depression (LTD). Stresses can cause changes by altering synapses through either LTP or LTD.
