Drosophila melanogaster has been extensively used in different types of research as a model organism due to its high reproductivity, short life span, and easy maintenance. In this paper, we examined drosophila as a model for ethanol toxicity in the male reproductive system. We developed three assays: The DAPI assay, sperm viability assay, and tubulin staining assay. The DAPI assay is a nuclear staining assay that assesses the effects of various ethanol concentrations in late spermiogenesis in which increased detrimental effects to the sperm bundles were observed. A Bioinformatics Methodology For The Annotation And Analysis Of Sperm Chromatin Remodeling Proteins In Sequenced Drosophila Species, Zain A. Alvi

Spermatogenesis is the process by which mature functional spermatozoa are formed, and is initiated in the stem cell niche of the testes. During the post-meiotic stage of spermatogenesis, spermiogenesis, transition proteins facilitate the transformation of chromatin from a histone-based nucleosome structure to a protamine-based nucleosome structure. This study is aimed at analyzing genomic, transcript, and protein sequences of transition proteins in 13 sequenced Drosophila species. The Drosophila melanogaster spermatid specific transition protein-like protein (Tpl94D) was used as the reference sequence in this study. The fruit fly Drosophila melanogaster is used extensively as a model for studying molecular, genetic and cellular aspects of human disease and physiology. Our lab has used D. melanogaster and related species to study the structure of the testis stem cell niche, as well as other aspects of spermatogenesis. We previously revealed a novel stem cell niche structure in D. pseudoobscura, a distant relative of D. melanogaster. The signaling center of the D. melanogaster stem cell niche has a well-characterized rosette arrangement of fasciclin-positive cells terms the “hub”. Spennatogenesis is a complex process that involves differentiation and morphological changes of diploid stem cells into highly specialized, haploid, motile germ cells. Many of the transformative processes involved in mammalian spermatogenesis are conserved in flies, and their study would be facilitated by the availability of a reliable in vitro culture system. Culture of Drosophila melanogaster spermatogenic cysts is limited in usefulness because of early degeneration of cysts, low yield of elongated spermatozoa, and rare occurrences of motile sperm. We have developed a culture system for studying fly spermatogenesis using isolated spermatogenic cysts from the testes of Drosophila pseudoobscura. This species …
