Heavy Metal Accumulation And Genotoxicity Indicator Capacity Of The Lichen Species Ramalina Pollinaria Collected From Around An Iron Steel Factory In Karabük, Turkey, Demet Cansaran Duman, Ersi̇n Altunkaynak, Emi̇ne Sümer Aras
Turkish Journal of Botany
In the present work, Ramalina pollinaria (Westr.) Ach. specimens collected from around an iron steel factory in Karabük were analyzed due to their heavy metal accumulation by atomic absorption spectrometry. The specimens were also evaluated for pollution-induced DNA damage by random amplified polymorphic DNA (RAPD) assay. Genomic template stabilities were calculated from the changes in RAPD profiles and compared with heavy metal content in R. pollinaria. The results obtained from chemical analysis suggest that R. pollinaria is a particularly suitable lichen species for the detection of air quality. Results of RAPD assay showed significant differences in band patterns in R. …

Uv-B Induces Cell Death In The Lichen Physcia Semipinnata (J.F.Gmel), Di̇lek Ünal, Yi̇ği̇t Uyanikgi̇l
Turkish Journal of Biology
We have examined the consequences of Ultraviolet-B (UV-B) irradiation in the thallus of the lichen Physcia semipinnata (J.F.Gmel) in terms of cell viability and apoptotic-like formation. UV-B induced oligonucleosomal DNA fragmentation was detected by TUNEL assay and this is the first study showing DNA fragmentation in thalli. The intensity of TUNEL-positive cells after exposure to UV-B at doses up to 95.9 J/cm2 was higher in the photobiont layer than the mycobiont layer.

Isolation Of Dna For Sequence Analysis From Herbarium Material Of Some Lichen Specimens, Sümer Aras, Demet Cansaran Duman
Isolation Of Dna For Sequence Analysis From Herbarium Material Of Some Lichen Specimens, Sümer Aras, Demet Cansaran Duman
Turkish Journal of Botany
An improved protocol for the isolation of DNA from herbarium material of some lichen specimens is described. The isolated DNA is suitable for PCR reactions for DNA sequence analysis. The hexadecyltrimethylammonium bromide (CTAB) based protocol defined in this study provides a number of advantages, mainly speed and reliability. In addition, different DNA extraction protocols were examined to determine the yield of DNA from the thallus of lichen specimens. The methods examined include a CTAB based protocol and a sodium dodecyl sulphate (SDS) based protocol. Although these procedures yielded DNA of suitable purity for PCR analysis, the improved protocol yielded DNA …

In this study, the levels of endogenous free, bound and total auxin (Indole-3-acetic acid, IAA), gibberellic acid (GA_3), abscisic acid (ABA) and cytokinin (zeatin) were examined in some species of mosses and lichens. For determination of the levels of these plant growth regulators, spectrophotometry was used. Our findings show that the mosses and lichens used in this study produce the plant growth regulators IAA, GA_3, ABA and zeatin.
