Investigating The Impact Of Transcription On Mutation Rates, Sarah Patterson

tRNA genes are highly transcribed and perform one of the most fundamental cellular functions. Although a universal pattern observed across all three domains of life is that highly transcribed genes tend to evolve slowly, tRNA genes have been shown previously to evolve rapidly. This rapid sequence evolution could result from relaxed selection, increased mutation rate, or a combination of both. Here, we use mutation-accumulation line sequencing data to show that tRNA genes accumulate more mutations than other gene types. Seminal plasma is commonly known to serve as a transport medium for sperm as it moves through the female reproductive tract for fertilization, however, more recent evidence demonstrates seminal plasma induces an expansive inflammatory response in the uterus. Murine models have found this inflammatory response is important for clearing pathogens and poor-quality sperm, eliciting the secretion of cytokines, chemokines, and embryokines that aid in embryo attachment and growth, placental angiogenesis, and blunting maternal immunity to the embryo. Softwood is more in demand than hardwood because it is used primarily in the US's largest wood-consuming industry, construction, resulting in increased importation of softwood annually to meet this demand. Hardwood, used for non-structural purposes like furniture and interior designs, is more abundant in US forests. However, some hardwood species are underutilized and undervalued. Cross-laminated timber (CLT) has increased the demand for softwood.

A study evaluated the mechanical and physical properties of three underutilized hardwood species (321 yellow poplar, 393 sweetgum, and 262 red oak specimens) for CLT industrial mat manufacturing. The results showed that red oak had a higher …

I investigated the effect of climate change in one of the few expanding markets in the world in Brazil. In the last decades the demand for pulpwood increased from zero to 11 million tons, leading to an expansion of 700 thousand hectares of Eucalyptus plantation. In 2024, a new mill will start operating increasing the consumption of wood fiber by 8.2 million tons (+49%). I used mathematical programming to investigate how different scenarios of productivity and land will affect the market. My results showed that around 946,000 hectares (+124%) of additional timberland will be necessary in the upcoming decades. The …

Estuarine habitats are considered nursery habitats for fishes and invertebrates, but oligohaline environments are critically understudied. Using a seine net and fyke nets, we sampled Back Bay, Mississippi (USA), a low salinity estuary, once a month for a year to explore the temporal use of the marsh and adjacent submerged landscape by nekton species. We also looked at whether there is evidence of habitat preference in the most numerous nekton species. We used a novel habitat usage index (HUI) to compare seine and fyke net catches of our top 10 species, and evaluated catch size in relation to maturation size. …

Zinc (Zn) has numerous functions and life sustaining processes depend on the presence of Zn within the body. Zinc sulfate is a common form of Zn supplemented in the beef industry. Inorganic sulfate based minerals have been associated with negative rumen effects, mainly, decreased rumen fermentation and protozoa numbers. Our studies concluded no difference in digestibility of three different forms of Zn (zinc glycinate, zinc sulfate, and zinc hydroxychloride). Although overall digestibility did not change, there were specific mineral concentration differences after steers consumed concentrate treatment for 10 days. Raptor nests on human-built structures represent a significant source of conflict as they can result in bird mortality, fires, structure damage, service distribution, or power outages when falling nest materials or animals connect with energized conductors. Power companies, such as the Tennessee Valley Authority (TVA), wish to mitigate these conflicts to avoid service disruptions. In this dissertation, I present my work towards understanding and mitigating the interactions between Ospreys (Pandion haliaetus) and human-made structures. Instrumental disparities and soil moisture are two of the key limitations in implementing spectroscopic techniques in the field. This study sought to address these challenges through two objectives. The first objective was to assess Visible-near infrared (VisNIR) and mid-infrared (MIR) spectroscopic approaches and explore the feasibility of transferring calibration models between laboratory and portable spectrometers. The second objective addressed the challenge of soil moisture and its impact on spectra. The portable spectrometers demonstrated comparable performance to their laboratory-based counterparts in both regions. One of the objectives of this research was to evaluate the need for an additional imidacloprid seed treatment on ThryvOn® (Bayer Crop Science®, St. Louis, MO) cotton to effectively manage tobacco thrips, Frankliniella fusca (Hinds). While evaluating thrips densities and damage ratings, ThryvOn cotton continued to display reduced damage and thrips populations than non-ThryvOn. Thrips populations and damage ratings were consistently reduced when incorporating a higher rate of imidacloprid, but they were not reduced enough to justify the higher rates of imidacloprid. Super Parents is a group-based, Extension-led parenting program developed to enhance the parent-child relationship. Implemented by trained Head Start staff using the train-the-trainer model, this effort is achieved by teaching positive parenting practices, increasing parents’ knowledge of child development, instructing parents in activities and exercises for enhancing their child’s executive function skills, and encouraging the use of mindfulness techniques in parenting. Volunteers’ Stewardship Action-Taking Experiences During The Covid-19 Pandemic Predicted By Their Motivation Toward Engagement And Ability To Teach Others, Suzanna R. Windon, Olga Buchko, Linda Falcone

The current study investigates the relationship between volunteer stewardship action-taking, motivation toward engagement in volunteer activities, and the ability to teach others during the COVID-19 pandemic. The final data set included 1,196 responses from the Penn State Extension Master Gardeners and Master Watershed volunteers, which provided a response rate of 39.9%. The results of this study showed that approximately 15.1% of the variation in volunteer stewardship action-taking experiences could be explained by volunteers’ motivation toward engagement in volunteer activities and their ability to teach others. Family and Consumer Sciences (FCS) and Extension professionals need to understand the lived experience of poverty because it affects every aspect of an individual’s life. Poverty is related to inadequate nutrition and food insecurity, lack of access to health care, insufficient child care, unsafe neighborhoods, lack of affordable housing, under-resourced schools, and a lower quality of life. Attitudes toward poverty vary widely among Americans and can be categorized as either internal/individual attributions (e.g., laziness, welfare dependency, etc.) or systemic/structural attributions (e.g., unemployment, inflation, etc.). Native bees are crucial for sustainable productivity in natural, agricultural, and urban ecosystems, but they are losing natural habitat spaces. Extension can facilitate community science programs to collect ecological data on native bee populations and support pollinator conservation. Native Bee Watch, an in-person community science program, transitioned to a hybrid format where volunteers received online training and support for conducting field-based data collection. This study focused on the professional development needs of Cooperative Extension agents and specialists with between four and seven years of experience, placing them roughly within the colleague career stage. Data were collected through focus groups and validated through member checking and the use of a modified World Café approach. A research team collaborated, increasing the reliability of the findings through intentional reflection in the development of the findings. Areas of competency strength and weakness vary for agents and specialists, and specific topic areas within competencies vary. Fifty-six percent of school-aged children participate in team sports, yet there is limited evidence on how participation in youth sports impacts the dietary behaviors of participants and their families. Our team surveyed parents and caregivers of children participating in recreation league youth sports (n = 178) to assess the foods and beverages offered as team snacks and factors that influence team snack choices. Juice was the most popular beverage, and crackers were the most popular snack reported by parents. Soil health-based agricultural management practices are widely promoted to improve soil structure, infiltration and reduce erosion. This study was conducted at two locations in North-Central Mississippi to evaluate the influence of different cover crop species and poultry litter on soil health that can impact crop production, climate change, and resilience. The results indicated that the cover crops showed a little effect on some soil health indicators compared to control treatment. The Forest Inventory and Analysis (FIA) program of the United States Department of Agriculture Forest Service collects forest inventory data that provide estimates with reasonable accuracy at the national scale. However, for smaller domains, these estimates are often not as accurate due to the small sample size. Small area estimation improves the accuracy of the estimates at smaller domains by relying on auxiliary information. Droughts are a common disturbance experienced by forest systems across the southeastern United States and are expected to increase in frequency in the near future. Few empirical studies have evaluated the response of individual tree resistance and resilience following silvicultural treatment in longleaf pine (Pinus palustris Mill.). This study evaluates the differences in drought responses between two forests during three different periods. The first period evaluated pretreatment (2000) conditions, while the second (2006), and third (2011) compared conditions after the implementation of five silvicultural treatments. Anthropogenic activities such as construction and agriculture directly affect its surrounding environments. Many studies have focused on how megafauna and flora are impacted by human activities, however, relatively little is known how new occupation of land affects smaller organisms such as insects and microbes. Additionally, other stressors such as predation can impact host physiology. This thesis aims to assess the efficacy of an emerging structured illumination reflectance imaging (SIRI) technique with machine learning approaches for assessing WS and microbial spoilage in broiler breast meat. Maximum size-density relationships (MSDR) are used to quantify differences across sites in the number of trees of a given size and species that can be supported per hectare. These relationships are important to managers who are trying to maximize basal area and wood volume. In my study, I examined MSDR across Alabama, Georgia, Louisiana, and Mississippi using US Forest Service, Forest Inventory and Analysis (FIA) data. I determined the impact of species-specific, specific gravity, functional traits, and environmental factors on MSDR using a quantile regression approach. Polyclonal plantings of Populus deltoides are expected to display increased site resource use, productivity, and tolerance to stress through plasticity changes leading to niche differentiation (i.e changes to crown/canopy structures). In the present study, P. deltoides Clones S7C8, 110412, and polyclonal plots were tested for differentially expressed genes and enriched biological pathways between planting schemes. Transcriptomic analysis of leaves revealed upregulation of an active growth gene and gene family members that play important roles in plant stress and stress tolerance in polyclonal plantings.
