Scanning Electron Microscopy
Cells of Paramecium caudatum were shock-frozen without pretreatment for cryoultramicrotomy and freeze-dried for subsequent X-ray microanalysis. Na, Mg, P, S, Cl, K, and Ca were detected in different amounts in several subcellular compartments. In particular, calcium was localized below the cell surface (pellicle). Trichocysts were found to contain significant amounts of Na in their base but not in the tip. Na, Mg, P, S, Cl, K, Ca were found in electron dense deposits within the lumen of the contractile vacuole. A small K concentration was found in the cytoplasm and in the mitochondria. The literature on scanning electron microscopy (SEM) pertaining to the retina has been surveyed and described.
The first two papers on SEM and retina appeared in 1969. Most of the earlier studies concentrated on descriptions [by SEM alone, or with light microscopy (LM) and transmission electron microscopy (TEM)] of the appearance of various retinae and retinal cells [fish, newt, primates, rodents and rabbits, bullfrog]; or embryology of the chick retina. Two papers dealt with retinal disease. In all there were 25 papers in SEM/retinal research before and in 1974. Since 1975 there have been 111 papers which have used SEM …

Earlier studies in the performance analysis of concurrency control algorithms in distributed database systems (DDBS) have assumed that the message transmission time between any two nodes of a network is constant. Hence they disregard the effect of communication subnet related parameters such as network traffic, network topology, the capacity of transmission channels and the size of messages transmitted. In this paper an analytical model is used to estimate the delays in transmission channels for a DDBS in a long haul network. The analysis shows that the constant transmission time assumption cannot be justified in many cases and that response time …

A Rapid Technique For Counting Cracks In Rocks, W. B. Durham, J. M. Beiriger, H. C. Weed
Scanning Electron Microscopy
Using a scanning electron microscope (SEM) and an image analyzer, we have developed a technique for counting and measuring cracks in rocks which is more efficient than traditional techniques in which an operator performs all image analysis functions. The key aspect of the technique is that black-on-white tracings of fresh cracks, which can be made rather rapidly by an operator, are measured and digitized by an image analyzer. The most time-consuming step in the process has now become the generation of SEM micrographs and pertinent chemical (mineralogical) information, not the quantification of crack structure. A temperature-dependent, two-component model, based on constant-temperature development data, was formulated which described the emergence of overwintered sorghum midges, Contarinia sorghicola (Coquillett) in the field. The first model component used a poikilotherm rate equation to predict emergence rates as a function of temperature. The second component distributed emerging adults over normalized time using a temperature- independent cumulative Weibull distribution. When coupled, components formed a temperature-dependent simulation model which described the spring emergence of overwintered midges from 1979 to 1982. The model tended to predict emergence before it actually occurred in the field. This discrepancy was corrected by incorporating a 1.27-cm …

The purpose of this study was to investigate plybond strength development of secondary fiber by the addition of the marine polymer chitosan.
The chitosan was applied to refined, lightly refined and unrefined pulp suspensions. Two-ply handsheets were produced with addition levels of .5, 1, 2, 3, and 4 percent chitosan based on fiber weight.
Results indicate that addition of chitosan to two-ply handsheets significantly increased plybond strength from 30 to 120 percent depending on furnish and degree of refining. It was also determined that maximum strength values of selected tests occurred at approximately the two percent addition level of chitosan. …

A study was made comparing papers dyed in a rosin/alum sized system to papers dyed in two alkaline sized systems. The alkaline sizes chosen were alkyl ketene dimer and alkenyl succinic anhydride. The dye types studied were acid, basic, anionic direct, and cationic direct. The degree of sizing was varied for each sizing type to see if the amount of sizing would effect dye retention. The papers dyed with anionic and cationic direct dyes and the basic dye were not significantly effected by sizing type of sizing degree. The direct dyes did give a lighter shade when the rosin size …

The response to periodic desiccation of periphyton communities developed in eddy environments or in areas exposed to variable, direct current was examined. Algal communities were incubated on clay tiles for between 2 and 12 weeks in the Colorado River, directly below Hoover Dam from October, 1983 - January, 1984. Mean daily discharge from the dam decreased over this period. To assess the influence of these discharge changes on community dynamics, samples collected over the first 12 weeks of the investigation were compared to samples of the same age collected over the final 8 weeks.

Paper made from TMP fibers offers good opacity and stiffness, but its use is limited because of its low strength. Chemical post-treatment of TMP offers possibilities of improving strength. Sulphonation of lignin appears to be the most economically feasible treatment. This thesis was oriented toward finding out if sulphonation of TMP fibers would improve the strength properties of TMP handsheets. Southern pine TMP fibers were treated with sodium sulfite (Na2SO3). The chemical concentration to be used was between 3-27% in alkali solution between pH 7-9. The temperature was between 90°-150°C. Pulp was consistency was 10%. Handsheets …

Since 1974, the use of ultraviolet-cured inks and coatings has become popular with the printing industry. Unfortunately for the deinking industry, removal of these inks is difficult using conventional methods in the U.S. Theory and laboratory work suggests these special inks may be removed using the flotation process.
It was discovered that the optimum flotation cell temperature for UV-cured inks is not significantly different than the optimum temperature for normal emulsifiable inks. However, optimum cell pH was found to be different between the two inks, and most effective at low pH for UV-cured ink.

This project concentrated on the development and subsequent results of an integrated deinking system. Although the chemistry and basic principles are quite different, a few European companies are successfully combining the two·systems by taking advantage of the differences.
Advantages of an integrated system over the conventional type included higher brightness, lower ash content and decreased sensitivity to particle size.
The experiment consisted of two different two-stage deinking systems. Each system was run twice; once with a sodium-precipitate chemistry and once with a calcium precipitate chemistry.

The objective of this study is to look at the effect of starch temperature on the emulsification of ASA. Historical information was limited, but it was discovered that an emulsion particle size range of 1-2 microns would give optimum size results.
A continuous emulsification apparatus was constructed using a glass venturi and power syringe to emulsify the starch and ASA. The basic conclusions drawn from the experiment were that the starch temperature did have an effect on the final sizing characteristics of the sheets. Refiner peroxide bleaching trials were carried out at the Abitibi-Price Research pilot plant using a Sunds-Defibrator CD300 pilot-scale refiner. Results show that, at the pilot scale, blowline addition of bleach liquor gives better brightness gains than addition in the refining zone. Using this technique, brightness gains equivalent to tower bleaching can be obtained when the bleaching parameters are properly controlled. The most important factors were post-refiner retention time and temperature, and the alkali charge in the bleach liquor. These three factors were shown to be highly interdependent. Refining pressure (temperature) was shown to have little effect on brightness gain in …

The objective of this laboratory study is to quantitatively analyze the distribution of adhesive during drying at different rates. The effect of binder migration on physical paper coating properties was also studied. This report reviews past laboratory studies on binder migration, and the factors that affect binder migration. The coating application was done on a laboratory keegan coater. Dried coating was physically removed using a surface grinder, and the grind offs were analyzed for latex concentration, to determine the degree of binder migration. A collaborative group for the study of multiple endocrine neoplasia type 2 (MEN-2) syndromes comprised of clinicians and laboratory scientists has been set up in the United Kingdom. Its aims are 1) to provide a basis for collaborative work on MEN-2; 2) to establish a register of patients; 3) and specifically to conduct studies aimed at defining the best policy for screening the families of apparently sporadic patients, to establish radioimmunoassays for family screening, and to identify large kindreds for genetic linkage studies using DNA polymorphisms.

In 19 patients from nine MEN-2A families, high-resolution C-banded chromosome studies have revealed a visible deletion within sub-band 20p12.2, yet no abnormality was observed in a 10th family. A deletion indistinguishable from that in MEN-2A was observed in five patients from three MEN-2B families but not in two other MEN-2B families. We found no abnormality in the entire karyotype of the four MEN-1 patients studied. These findings suggest that the mutation in most MEN-2 patients is a visible deletion in the short arm of chromosome 20.

Through nationwide surveys, we collected and analyzed 242 patients of medullary thyroid carcinoma (MTC). Included were 40 patients with multiple endocrine neoplasia type 2A (MEN-2A), six patients with MEN-2B, and 36 patients with only MTC having a positive family history (82 total patients in the hereditary group). Ten-year survival rates were 81.5% for all cases, and 97.5% and 76.1% for the hereditary and the sporadic group, respectively. Epinephrine/norepinephrine ratio in the urine was found to be a good indicator of the adrenomedullary hyperfunction in patients with hereditary MTC. A study designed to identify the location of a gene for multiple endocrine neoplasia type 2 (MEN-2) is being performed using a single large family segregating for MEN-2A. Here we report our initial results using a set of DNA restriction fragment length polymorphisms spanning a 40 cM segment of the short arm of human chromosome 11. The locus for MEN-2 in this family is excluded from this entire region which includes an oncogene, c-Ha-ras-1, two hormone loci, insulin and parathyroid, and the five hemoglobin genes in the beta hemoglobin gene duster. Using recombinant DNA techniques, we analyzed the structure of human calcitonin (CT)-encoding DNA. The sequence of nucleotides derived from the messenger RNA (mRNA) predict the amino acid sequence of the entire human CT precursor protein. The complete CT gene could be isolated from a DNA library of the human genome. The CT precursor encoding sequences (exons) as well as the noncoding intervening sequences (introns) could be identified. Another sequence, called human calcitonin gene-related peptide (hCCRP), could be located near this CT-encoding DNA. We reviewed the scintigraphic images made after injections of 1-131 metaiodobenzylguanidine (MIBG) or 1-123 MIBG in patients with multiple endocrine neoplasia (MEN) types 2A and 2B. The information we obtained was applied to three questions about the treatment of pheochromocytoma in patients affected with these syndromes. Our first question "When should adrenal gland(s) be removed?" was not directly answered. However, adrenalectomy generally should not be contemplated unless distinct abnormalities are present in the scintigraphic images. With experience it may be possible to approximate, from the stage of pheochromocytoma depicted by scintigraphy, how many years will elapse before symptoms or hypertension …
