The Use Of Bacteriophages For Food Safety, Lorraine Endersen, Aidan Coffey Dec 2020

The Use Of Bacteriophages For Food Safety, Lorraine Endersen, Aidan Coffey

Department of Biological Sciences Publications

The search for natural biocontrol agents that allow the production of foods that are safe for human consumption and do not impact the taste, texture, and nutritional quality of the food, is a constant challenge for diverse food industries worldwide, particularly as the human population continues to rise globally, and multiple antibiotic resistance in pathogenic bacteria is increasingly prevalent. Bacteriophages (phages), the naturally occurring predators of bacteria, are harmless to humans and animals and are ubiquitous in the environment — and as such, have been recognised as promising antimicrobial agents to help control specific bacterial pathogens in food production. This …

The development of reliable, mixed-culture biotechnological processes hinges on understanding how microbial ecosystems respond to disturbances. Here we reveal extensive phenotypic plasticity and niche complementarity in oleaginous microbial populations from a biological wastewater treatment plant. We perform meta-omics analyses (metagenomics, metatranscriptomics, metaproteomics and metabolomics) on in situ samples over 14 months at weekly intervals. Based on 1,364 de novo metagenome-assembled genomes, we uncover four distinct fundamental niche types. Throughout the time-series, we observe a major, transient shift in community structure, coinciding with substrate availability changes. Functional omics data reveals extensive variation in gene expression and substrate usage amongst community members. …

Biology Faculty Publications and Presentations

Elevation is an important determinant of ecological community composition. It integrates several abiotic features and leads to strong, repeatable patterns of community structure, including changes in the abundance and richness of numerous taxa. However, the influence of elevational gradients on microbes is understudied relative to plants and animals. To compare the influence of elevation on multiple taxa simultaneously, we sampled phytotelm communities within a tropical pitcher plant (Nepenthes mindanaoensis) along a gradient from 400 to 1200 m a.s.l. We use a combination of metabarcoding and physical counts to assess diversity and richness of bacteria, micro-eukaryotes, and arthropods, and …

Life Sciences Faculty Research

The Atlantic horseshoe crab (Limulus polyphemus) is a common marine aquarium species and model organism for research. There is potential monetary and conservation value in developing a stable captive population of horseshoe crabs, however, one major impediment to achieving captivity is a lack of knowledge regarding captive diseases. We utilized 16S rRNA gene amplicon sequencing to track changes in the microbiomes of four body locations in three wild-caught (tracked over 14 months in captivity) and three tank-acclimated (>2 years in captivity) adult L. polyphemus in a touch tank at Shark Reef Aquarium at Mandalay Bay in Las Vegas, NV. …

Microbiomes Of Caribbean Octocorals Vary Over Time But Are Resistant To Environmental Change, Mark Mccauley, Colin R. Jackson, Tamar Goulet

Faculty and Student Publications

The bacterial microbiome is an essential component of many corals, although knowledge of the microbiomes in scleractinian corals far exceeds that for octocorals. This study characterized the bacterial communities present in shallow water Caribbean gorgonian octocorals over time and space, in addition to determining the bacterial assemblages in gorgonians exposed to environmental perturbations. We found that seven shallow water Caribbean gorgonian species maintained distinct microbiomes and predominantly harbored two bacterial genera, Mycoplasma and Endozoicomonas. Representatives of these taxa accounted for over 70% of the sequences recovered, made up the three most common operational taxonomic units (OTUs), and were present in …
