Viruses encoding a replication-associated protein (Rep) within a covalently closed, single-stranded (ss)DNA genome are among the smallest viruses known to infect eukaryotic organisms, including economically valuable agricultural crops and livestock. Although circular Rep-encoding ssDNA (CRESS DNA) viruses are a widespread group for which our knowledge is rapidly expanding, biased sampling toward vertebrates and land plants has limited our understanding of their diversity and evolution. An important question in many biological applications, is to estimate or classify gene activity states (active or inactive) based on genome-wide transcriptomics data. Recently, we proposed a Bayesian method, titled MultiMM, which showed superior results compared to existing methods. In short, MultiMM performed better than existing methods on both simulated and real gene expression data, confirming well-known biological results and yielding better agreement with fluxomics data. Despite these promising results, MultiMM has numerous limitations. We investigated alternatives to whole blood for blood feeding of mosquitoes with a focus on improved stability and compatibility with mass rearing programs. In contrast to whole blood, an artificial blood diet of ATP-supplemented plasma was effective in maintaining mosquito populations and was compatible with storage for extended periods refrigerated, frozen, and as a lyophilized powder. The plasma ATP diet supported rearing of both Anopheles and Aedes mosquitoes. It was also effective in rearing Wolbachia-infected Aedes mosquitoes, suggesting compatibility with vector control efforts.

Background: Juvenile hormones (JH) and ecdysteroids control postembryonic development in insects. They serve as valuable targets for pest management. Hence, understanding the molecular mechanisms of their action is of crucial importance. CREB-binding protein (CBP) is a universal transcriptional co-regulator. It controls the expression of several genes including those from hormone signaling pathways through co-activation of many transcription factors. However, the role of CBP during postembryonic development in insects is not well understood. Therefore, we have studied the role of CBP in postembryonic development in Tribolium, a model coleopteran insect.

Leishmaniasis is a parasitic infection that afflicts approximately 12 million people worldwide. There are several limitations to the approved drug therapies for leishmaniasis, including moderate to severe toxicity, growing drug resistance, and the need for extended dosing. Moreover, miltefosine is currently the only orally available drug therapy for this infection. We addressed the pressing need for new therapies by pursuing a two-step phenotypic screen to discover novel, potent, and orally bioavailable antileishmanials. The contribution of high fructose corn syrup (HFCS) to metabolic disorder and obesity, independent of high fat, energy-rich diets, is controversial. While high-fat diets are widely accepted as a rodent model of diet-induced obesity (DIO) and metabolic disorder, the value of HFCS alone as a rodent model of DIO is unclear. Impaired dopamine function is associated with obesity and high fat diet, but the effect of HFCS on the dopamine system has not been investigated. Celidophylla albimacula Saussure and Pictet, 1898 (Orthoptera: Tettigoniidae) is reported for the first time from Panama. The specimen was collected in the Turega Water Reserve in the province of Coclé. Celidophylla albimacula had only been previously reported for Costa Rica and Nicaragua.

Species of Mortoniella are revised for the northern and Andean part of the South American continent, including the countries of Bolivia, Peru, Ecuador, Colombia, Venezuela, and Guyana. All previously described species from the region are reillustrated and redescribed, except for Mortoniella santiaga Sykora, 1999 and M. quinuas Harper and Turcotte, 1985, whose types could not be located, and M. tranquilla Martynov, 1912, whose type is based on a female specimen and thus is currently unidentifiable. We measured total soil CO2 efflux (RS) and efflux from the forest floor layers (RFF) in red pine (Pinus resinosa Ait.) stands of different ages to examine relationships between stand age and belowground C cycling. Soil temperature and RS were often lower in a 31-year-old stand (Y31) than in 9-year-old (Y9), 61-year-old (Y61), or 123-year-old (Y123) stands. This pattern was most apparent during warm summer months, but there were no consistent differences in RFF among different-aged stands. RFF represented an average of 4–13% of total soil respiration, and forest floor removal increased …

A new cryptic species, Aleochara (Xenochara) castaneimarmotae Klimaszewski, Webster, and Brunke, new species (Coleoptera: Staphylinidae: Aleocharinae), associated with Marmota monax (Linnaeus) burrows and caves in eastern North America, is described and illustrated. A key to Canadian species of subgenus Xenochara Mulsant and Rey and revised distributions of the taxonomically difficult fumata species group are provided. Aleochara quadrata Sharp is recorded from Washington and Oregon for the first time.

Many nomenclatural changes are implemented in the beetle families Georissidae, Histeridae, Hydraenidae, Hydrochidae, Hydrophilidae, Ptiliidae, Leiodidae and especially Staphylinidae, of the beetle series Staphyliniformia (Coleoptera), in preparation for making a world catalog of this group available online. Limited taxonomic changes are also made in the staphylinid subfamilies Osoriinae and Staphylininae.
At the level of family-group taxa, Article 29.4 of the current (1999) Zoological Code is reviewed and the original spellings of two tribal names, Nymphisterini Tishechkin (Histeridae) and Cryptonotopsisini Pace (Staphylinidae), are resurrected. This paper revises the genus Ganelius Benesh, which is endemic to Madagascar, in the stag beetle tribe Figulini Burmeister (Coleoptera: Lucanidae: Lucaninae). Species in the genus are striking in their highly sexually dimorphic mandibles, a character that rarely occurs in the tribe. The genus was previously comprised of four species, but three of these names were problematic or questionably applied. A lectotype is designated for G. madagascariensis Laporte, a neotype is designated for Ganelius oberndorferi Nonfried, and Nigidius passaliformis Benesh is newly synonymized with G. oberndorferi. The tribe Zolini (Carabidae: Trechinae) is revised for New Zealand. Two subtribes, five genera, and fortyeight species are recognized. The presence of the subtribe Merizodina is confirmed for New Zealand; it includes three genera (Maungazolus n. gen., Pseudoopterus Csiki, 1928, and Synteratus Broun, 1909). The subtribe Zolina contains two genera (Oopterus Guérin-Méneville, 1841 and Zolus Sharp, 1886).

Developmental biology (including embryology) is proposed as "the stem cell of biological disciplines.” Genetics, cell biology, oncology, immunology, evolutionary mechanisms, neurobiology, and systems biology each has its ancestry in developmental biology. Moreover, developmental biology continues to roll on, budding off more disciplines, while retaining its own identity. While its descendant disciplines differentiate into sciences with a restricted set of paradigms, examples, and techniques, developmental biology remains vigorous, pluripotent, and relatively undifferentiated. In many disciplines, especially in evolutionary biology and oncology, the developmental perspective is being reasserted as an important research program.

Endocytosis is key to fibrinogen (Fg) uptake, trafficking of integrins (αIIbβ3, αvβ3), and purinergic receptors (P2Y1, P2Y12), and thus normal platelet function. However, the molecular machinery required and possible trafficking routes are still ill-defined. To further identify elements of the platelet endocytic machinery, we examined the role of a vesicle-residing, soluble N-ethylmaleimide factor attachment protein receptor (v-SNARE) called cellubrevin/vesicle-associated membrane protein-3 (VAMP-3) in platelet function. Although not required for normal platelet exocytosis or hemostasis, VAMP-3−/− mice had less platelet-associated Fg, indicating a defect in Fg uptake/storage. Other granule …

This unique case series and review of literature suggests that immune checkpoint inhibitors may have clinical activity in neuroendocrine tumors.
Objective: Summarize advances of immuno-oncology in neuroendocrine tumors with the help of a case series.
Design: Case series and review of literature.
Intervention or Exposure: The patients were treated with immune checkpoint inhibitors (pembrolizumab or nivolumab).
Main Outcome(s) and Measures(s): Life expectancy, quality of life, disease progression.
Results: Maximum durable response of 16 months in one of the patients so far. All patients showed improvement in quality of life before disease progression. Two out of four are still on therapy. …

Storage roots of cassava (Manihot esculenta Crantz), a major subsistence crop of sub-Saharan Africa, are calorie rich but deficient in essential micronutrients, including provitamin A β-carotene. In this study, β-carotene concentrations in cassava storage roots were enhanced by coexpression of transgenes for deoxy-d-xylulose-5-phosphate synthase (DXS) and bacterial phytoene synthase (crtB), mediated by the patatin-type 1 promoter. Storage roots harvested from field-grown plants accumulated carotenoids to ≤50 lg/g DW, 15- to 20-fold increases relative to roots from nontransgenic plants. Geographic differences in the age structure of 4 populations of ocean quahogs Arctica islandica throughout the range of the stock within the US exclusive economic zone were examined. The ages of animals fully recruited to the commercial fishery (≥80 mm shell length) were estimated using annual growth lines in the hinge plate. The observed age frequency from each site was used to develop an age−length key enabling reconstruction of the population age frequency for the site. Neurofeedback relying on functional magnetic resonance imaging (fMRI-nf) heralds new prospects for self-regulating brain and behavior. Here we provide the first comprehensive review of the fMRI-nf literature and the first systematic database of fMRI-nf findings. We synthesize information from 99 fMRI-nf experiments—the bulk of currently available data. The vast majority of fMRI-nf findings suggest that self-regulation of specific brain signatures seems viable; however, replication of concomitant behavioral outcomes remains sparse. To disentangle placebo influences and establish the specific effects of neurofeedback, we highlight the need for double-blind placebo-controlled studies alongside rigorous and standardized statistical analyses. Vocal traits can be sexually selected to reflect male quality, but may also evolve to serve additional signaling functions. We used a long-term dataset to examine the signaling potential of song in dimorphic white-throated sparrows (Zonotrichia albicollis). We investigated whether song conveys multifaceted information about the vocalizing individual, including fitness, species identity, individual identity, and morph. We also evaluated whether song traits correlate differently with fitness in the two morphs, as the more promiscuous strategy of white, relative to tan, morph males might impose stronger sexual selection. Tissue factor pathway inhibitor α (TFPIα) inhibits prothrombinase, the thrombin-generating complex of factor Xa (FXa) and factor Va (FVa), during the initiation of coagulation. This inhibition requires binding of a conserved basic region within TFPIα to a conserved acidic region in FXa-activated and platelet-released FVa. In this study, the contribution of interactions between TFPIα and the FXa active site and FVa heavy chain to prothrombinase inhibition were examined to further define the inhibitory biochemistry. Aneuploidy disrupts cellular homeostasis. However, the molecular mechanisms underlying the physiological responses and adaptation to aneuploidy are not well understood. Deciphering these mechanisms is important because aneuploidy is associated with diseases, including intellectual disability and cancer. Although tumors and mammalian aneuploid cells, including several cancer cell lines, show altered levels of sphingolipids, the role of sphingolipids in aneuploidy remains unknown. Here, we show that ceramides and long-chain bases, sphingolipid molecules that slow proliferation and promote survival, are increased by aneuploidy. Sphingolipid levels are tightly linked to serine synthesis, and inhibiting either serine or sphingolipid synthesis can specifically impair the fitness …
