The continued rise of antibiotic-resistant bacteria, such as MRSA (methicillin-resistant Staphylococcus aureus), has made the discovery of novel antibiotics critical. While bacteriostatic and bactericidal antibiotics inevitably exert evolutionary pressure on bacteria to develop resistance, small molecules that target mechanisms of bacterial virulence present a promising alternative for treatment. Sortase A (Srt A), a cysteine protease of S. aureus, promotes bacterial virulence by covalently attaching proteins such as pilin to the bacterial surface, enabling bacterial adhesion to mammalian cells. Membrane transporter proteins play the vital role of moving compounds in and out of the cell and are essential for all living organisms. ATP Binding Cassette (ABC) family transporters function both as importers and exporters in prokaryotes. MetNI is an E. coli Type I ABC transporter responsible for the uptake of methionine into the cytosol from the cell periplasmic space through the cell membrane to maintain intracellular methionine pools. ABC transporters, like other membrane proteins, are most often mechanistically and structurally studied in vitro, solubilized by detergents. As the global populations grow, the need for stronger agricultural resilience in areas with high food insecurity like Sub-Saharan Africa (SSA) is a must. Crop failure due to climate related events has increased substantially over the past decade, raising the risks to vulnerable groups like subsistence farmers. Understanding the effects of climate variability on farmers and their crops is a pivotal step towards increasing resilience and productivity. In this paper I approach this question using a Linear Probability Fixed-effects Model on Tanzanian Panel Survey data with monthly temperature and precipitation for years 2008 to 2013. Play behavior is ubiquitous among young mammals and has a variety of important influences on early development. I evaluated possible influences of social play behavior on the development of temperament in juvenile Belding’s ground squirrels (Urocitellus beldingi), using the caution/boldness spectrum as a measure of temperament. I disrupted the play behavior of juvenile U. beldingi by walking toward juveniles at play until the play interactions stopped. I used undisrupted juveniles and juveniles whose non-play behavior was disrupted at regular intervals as controls.
