Black ash (wikpiyik/Fraxinus nigra) and sweetgrass (suwitokolasol/Anthoxanthum nitens) are two culturally important species to Wabanaki (Passamaquoddy, Penobscot, Maliseet, Mi’kmaq, and Abenaki) people, and to many other Native American tribes across the species’ ranges. Wabanaki relationships with black ash and sweetgrass include creation stories, generational stewardship practices, important economic markets, and cultural identity. Land use patterns, changes in access, invasive species, and climate change are negatively affecting both the health of black ash and sweetgrass and Wabanaki people’s relationship to these species. Streptococcus agalactiae (Group B Streptococcus or GBS), a Gram-positive bacterium, and Candida albicans, a polymorphic fungus, are commensal microbes in most of the population they colonize but are also capable of causing severe and sometimes fatal infections in certain patient groups. Both organisms share similarities including the colonization the same tissue environments and causing infections in specific patient groups including those who are newborn, pregnant, suffering from chronic conditions like diabetes and HIV, as well as elderly patients. Previous research discovered that GBS and C. albicans can synergize to enhance the colonization of GBS in the bladders of mice, but …

In recent times, plastic has become a highly favored choice for packaging due to its exceptional microbial, damage, and water-resistant properties. However, the alarming rise in plastic usage has led to adverse environmental pollution. This study aims to develop innovative food packaging solutions using renewable and compostable cellulose nanofibrils (CNFs). This thesis comprises two related studies on the barrier performance of novel food packaging materials, including oxygen barrier and oil/grease barrier properties. In an era of increasing natural disturbances, successful tree regeneration has grown more difficult to achieve. Salvage logging, a common management response to disturbance, may further impede regeneration success, although published literature currently remains inconclusive. In 2013, a rare tornado in northcentral Maine, USA, and subsequent salvage operation created three clear ‘treatments’ for evaluation of post-disturbance regeneration: blowdown, blowdown followed by salvage logging and an undisturbed control. In the summers of 2022 and 2023, (nine and ten) years post-tornado, we revisited this site to examine regeneration outcomes.

Spruce budworm (Choristoneura fumiferana; SBW) outbreaks are cyclically occurring phenomena in the northeastern USA and neighboring Canadian provinces. These outbreaks are often of landscape level causing impaired growth and mortality of the host species namely spruce (Picea sp.) and balsam fir (Abies balsamea (L.) Mill.). Acknowledging the recent SBW outbreak in Canadian provinces like Quebec and New Brunswick neighboring the state of Maine, our study devised comprehensive techniques to assess the susceptibility of Maine forests to SBW attack. Forests provide numerous ecological and socio-economic benefits, yet climate change is creating novel and extreme conditions that threaten forests and disrupt traditional management practices. To address future uncertainty about how to manage forests amid a rapidly changing climate, researchers have developed adaptive management strategies that move away from using historical ecological baselines as management goals. Adipogenesis is regulated by the coordinated activity of adipogenic transcription factors, including PPAR-gamma (PPARG) and C/EBP alpha (CEBPA). Thus, dysregulated adipogenesis predisposes adipose tissues to adipocyte hypertrophy and hyperplasia. We have previously reported that mice possessing a homozygous null gene mutation in collagen triple helix repeat-containing protein 1 (CTHRC1) have increased adiposity compared to wildtype mice, supporting the concept that CTHRC1 regulates body composition. Herein, we investigated the anti-adipogenic activity of CTHRC1. Using 3T3-L1 preadipocytes, we showed significantly reduced adipogenic differentiation in the presence of CTHRC1 commensurate to marked suppression of Cebpa and Pparg gene expression. Horse behavior in pasture and grazing environments remains understudied, despite the substantial domestic horse population in the United States. This paper explores the utilization of Global Positioning System (GPS) technology to detect equine behavior, specifically focusing on grazing behaviors. By analyzing GPS data from (n=9) Standardbred horses, this study aims to establish the capabilities and accuracy of the Columbus P-1 data logger as a low-cost GPs unit for equine research, without the use of accelerometers. Through the data provided by the GPS unit, a model to distinguish grazing, resting, drinking, walking, trotting, and running was developed with satisfactory detection rates. …

Polymicrobial infections pose a significant challenge in the field of medicine due to our incomplete understanding of how pathogens interact during infection and how these interactions impact the effectiveness of drug treatments. Among the opportunistic pathogens, Candida albicans and Pseudomonas aeruginosa stand out, as they are often found in similar infection sites, such as burn wounds, the lungs of patients with cystic fibrosis and those on mechanical ventilation. Candida albicans is an opportunistic fungus responsible for invasive candidiasis, a condition associated with a high mortality rate of 40% in hospitals. Assessing avian health is not only important for the conservation and management of wild birds and the recreational economy, but also for the management of infectious diseases that threaten public health and agriculture. Sous-vide cooking is a thermal processing method in which a raw food is vacuum sealed in a pouch and then placed into water below 100°C for a controlled amount of time. It is particularly good for cooking meats because of its precise control over temperature and time, which results in an ideal food texture for consumers. This method also prevents the food from coming in contact with oxygen, which can lead to spoilage that reduces product quality and shelf life. To increase understanding of both the adoption rate and in-state manufacturing of mass timber In Maine, the 131st Legislature and Governor Mills passed LD 881, a resolve directing a study of the barriers facing cross-laminated timber In Maine and provide recommendations to promote their use in construction. This study was developed in response to the resolve. The study engaged 108 unique participants to define available training, education, and experiences across the stakeholders throughout the building lifecycle process in the state.
From this information, the following five recommendations are proposed:

Throughout the last decade, the oyster aquaculture industry in Maine has boomed, with 6 million pounds of oysters being harvested in 2021, about the amount that was harvested in 2016 (Cough, 2022). According to the founders of the Maine Oyster Trail, there are over 150 oyster farms currently operating on Maine’s coast, which is about twice as many as there were in 2016 (Maine Sea Grant). Seventy-five percent of coral reefs globally face crisis due to anthropogenic disturbances, prompting heightened global coral restoration initiatives to preserve these vital ecosystems. Various regions employ diverse active coral restoration methodologies, including coral gardening, transplantation, micro-fragmentation, artificial reefs, and sexual propagation. Of these methods, coral gardening stands out as one of the most common and highly successful methods, alongside widespread transplantation practices. Restoration efforts predominantly focus on acroporids due to their relatively rapid growth and asexual fragmentation; however, a diverse range of coral species, including large, slow-growing varieties, is also employed in these endeavors. Past research has found that female Drosophila generally decrease their reproductive output in response to exposure to predators, including parasitoid wasps. However, no studies on behavioral changes induced by the endoparasitic wasp Ganaspis brasiliensis have been documented in the literature. G. brasiliensis has been identified as a biocontrol agent candidate against D. suzukii, with field trials currently underway across the contiguous United States. In this thesis, two experiments were performed: a behavioral observation assay measuring reproductive behaviors and an oviposition assay, a measure of reproductive activity. Female D. suzukii exposed to G. brasiliensis were observed to have depressed oviposition, producing …

AquEOUS Fellowship. The inaugural summer of this new USDA fellowship program welcomed six undergraduate students to Orono from UMaine and four other U.S. colleges and universities. Over the course of 10 weeks, they approached aquaculture projects with "two-eyed seeing," incorporating local indigenous perspectives. The main objective of this thesis is to improve the understanding and awareness of methodologies to decrease phytoestrogens in conserved legumes without sacrificing forage nutritive value. In chapter 1, we discussed the main factors influencing each stage of hay production and our current understanding of the hay microbiome dynamics. The primary objective of haymaking is to dry forage enough (80-85% DM) to inhibit the growth of undesirable microbes and halt residual plant enzymatic activity that causes nutrient losses. The Interleukin-17 (IL17) family is a group of cytokines implicated in the etiology of several inflammatory diseases. Interleukin-17 receptor D (IL17RD), also known as Sef (similar expression to fibroblast growth factor), belonging to the family of IL17 receptors, has been shown to modulate IL17A-associated inflammatory phenotypes. The objective of this study was to test the hypothesis that IL17RD promotes endothelial cell activation and consequent monocyte adhesion. We utilized primary human aortic endothelial cells and demonstrated that RNAi targeting of IL17RD suppressed transcript levels by 83% compared to non-targeted controls. Further, RNAi knockdown of IL17RD decreased the adhesion of THP-1 cells …

Ocean warming and pollution are two detrimental anthropogenic factors causing rapid marine ecosystem degradation recorded in the past decades. These factors alter the marine environment intolerable for many marine species, forcing them to either adapt or shift their contemporary habitat ranges to reduce the extinction risk embedded with environmental degradation. Estimating marine species’ habitat range shifts, and their potential for developing adaptive mechanisms are critical for ecosystem conservation and management, human health risk assessment, and climate change vulnerability assessments. In Maine, the decline of cave dwelling bats has been caused by the fungal disease commonly known as white-nose syndrome discovered in 2006. Northern long-eared bats (Myotis septentrionalis), little brown (Myotis lucifigus) andeastern tricolored bats (Perimyotis subflavus) have experienced 95% or greater population declines, and eastern small-footed (Myotis leibii) and big brown bats (Eptesicus fuscus) have also declined. Long-Term Influence Of Commercial Thinning On Spruce-Fir Forests, Bishnu H. Wagle Mr.

This study investigates long-term influences of pre-commercial thinning (PCT) and commercial thinning (CT) treatments in spruce-fir forests, a key forest type in northeastern North America. Utilizing repeated measurements from replicated experimental research sites (n=15) in Maine, both stand- and tree-level responses to contrasting CT with and without prior PCT (NoPCT) have been quantified. In addition, an in-depth economic analysis, exploring the impact of different thinning treatments, associated costs, merchantability specifications, varying timber and carbon prices, and discount rates, on net present value (NPV) has been carried out using both measured and projected data.
