Excitement is a problem when conducting endocrine tests on fractious horses. Epinephrine (EPI), for example, was shown to obliterate the insulin-induced decrease in blood glucose concentrations. Sedation may be a solution; however, perturbation of results may preclude useful information. Six insulin sensitive and six insensitive mares were used in a replicated 3 x 3 Latin square design to determine the pituitary hormonal responses (compared to vehicle) to sulpiride and thyrotropin-releasing hormone (TRH), two compounds commonly used to diagnose pituitary pars intermedia dysfunction (PPID) in horses. Mares were classified as insulin sensitive or insensitive by their previous glucose responses to direct injection of human recombinant insulin. Treatment days were February 25 and March 10 and 24, 2012. Treatments were sulpiride (racemic mixture, 0.01 mg/kg BW), TRH (0.002 mg/kg BW), and vehicle (saline, 0.01 mL/kg BW) administered intravenously. Blood samples were …

Two experiments assessed the potential of altrenogest, an artificial progestogen, and deslorelin acetate, a gonadotropin-releasing hormone (GnRH) agonist, for suppressing estrus and ovulation in cyclic mares. In the first experiment, mares were administered a luteolytic dose of prostaglandin-F2á on d 6 of diestrus, and were then treated with 1 of 3 formulations of altrenogest in slow-release vehicle (6 mares/group): 1) Biorelease altrenogest LA150 (225 mg total as a 1.5 mL injection); 2) Biorelease altrenogest LA225 (225 mg total as a 1.0 mL injection); or 3) Biorelease altrenogest LA225 (450 mg total as a 2.0 mL injection). Mares of poor body condition have low plasma leptin concentrations, while obese mares have the highest concentrations. Leptin is a primary signal of body condition to the brain in other species; therefore, low leptin concentrations in thin mares could contribute to their extended anovulatory period in winter compared to obese mares. The current experiment was designed to determine whether recombinant equine leptin, administered to seasonally anovulatory mares, would induce ovarian activity and ovulation in the winter. Reproductive Characteristics Of High Body Condition Mares With High Versus Low Leptin Concentrations, Cara A. Waller

Two experiments were performed to discover what, if any, reproductive differences exist in high body condition (BCS) mares with varying concentrations of leptin. Previous research showed that mares with high body condition scores can have a range of leptin levels, from very low (<5 ng/mL) to very high (>10 ng/mL). Earlier results indicated that most mares with high body condition scores maintain estrous cycles or show significant follicular activity during the winter. Among these high BCS mares, about 30% of them exhibit hyperleptinemia and hyperinsulinemia. An experiment was conducted to determine the effects of dihydrotestosterone (DHT) and estradiol benzoate (EB) given simultaneously on concentrations of FSH and LH and the associated follicular activity, and to monitor the subsequent recovery of the gonadotropes and follicular growth following treatment with either DHT or EB. Twelve mares were assigned to daily treatments of DHT (150 Fg/kg BW) plus EB (22 Fg/kg BW) from d 0 through 30 (Phase I).
