The human hemizona has been demonstrated to select spermatozoa with good motility, normal morphology, and the capacity to undergo the zona-induced acrosomal reaction. The studies conducted here are directed at using the human hemizona to investigate two key questions: (1) whether human X-chromosome bearing spermatozoa (X-sperm) and Y-chromosome bearing spermatozoa (Y-sperm) differ in their functional survival time (assessed by their capacity to bind to the human zona pellucida) after prolonged in vitro incubation, and (2) whether hemizona-bound spermatozoa have a reduced aneuploidy rate compared to unbound (and therefore, unselected) spermatozoa?

Despite adequate hormonal stimulation, oocytes collected for the purpose of in vitro fertilization and embryo transfer display several levels of nuclear maturity. Preovulatory or mature oocytes, technically those that are Metaphase I or II, are inseminated shortly after aspiration and assessed for fertilization the following day. Prophase I oocytes, also called germinal vesicle-bearing or immature oocytes, require a 24-36 hour period in culture before being exposed to spermatozoa. During this time, the majority of Prophase I oocytes complete nuclear maturation in vitro, progressing from germinal vesicle breakdown through first polar body extrusion. Cancers of the oral cavity and pharynx have increased throughout the decades. Since oral cancer represents a small percentage of the total cancers in the United States, it is often overlooked. Though this is the case, the results of this cancer can be devastating. To address this issue, an Oral Cancer Screening was organized on August 14, 1996 at the West Michigan Cancer Center in Kalamazoo. Study Of The Hiv-1 Gene Regulatory Proteins Tat And Rev By The Two-Hybrid System In Yeast, David Allen Elkins

Tat and Rev, small proteins encoded by the Human Immunodeficiency Virus type 1 (HIV-1), regulate its pattern of gene expression within infected cells. Tat increases transcription from the integrated pro virus by approximately a thousand-fold; Rev effects a shift in the pattern of splicing of the viral mRNA. Both are likely to interact with cellular proteins in executing their respective functions. Tat has additional activities (including neurotoxicity and inhibition of lymphocyte activation) which may be extraneous to its viral functions.
In order to identify cellular proteins interacting with Tat and Rev, the genes for both were cloned into plasmid vectors …

Staphylococcus aureus chromosomal DNA from 125 methicillin-resistant and 10 methicillin-sensitive clinical isolates was digested with rare-cutting restriction endonucleases and subjected to pulsed-field gel electrophoresis (PFGE). Thirty-nine distinct genomic macrorestriction patterns (GPs) were identified using SmaI; 29 of these patterns are from methicillin-resistant S. aureus (MRSA).
A dendrogram showing percent similarity among the patterns was constructed which revealed the considerable genomic diversity of the collection even though the isolates had been obtained during outbreaks. The majority of the MRSA formed one broad group with two outbreak subclasses. However, 8 of the 29 MRSA GPs (28%) were diverse.

Homology Among The Autosomal Chromosomes Of Boophilus Annulatus (Say) And B. Microplus (Canestrini), Bonnie S. Gunn

Autosomes of Boophilus annulatus and B. microplus were compared using C- and G-bands to establish the degree of homology. Karyotypes of both species consisted of 20 autosomes and an XX:XO sex determination system with the X being the largest chromosome. All chromosomes of B. annulatus were acrocentric with heterochromatin limited to centromeric regions. The B. microplus karyotype was acrocentric with one band of noncentromeric heterochromatin occurring in three chromosome pairs. Interspecific comparisons indicated seven pairs of G-band homologous autosomes and three G-band homologous pairs when added interstitial heterochromatin was considered. The purpose of the project was to assess the effects of inbreeding on the genetic constitution of two Pakistani bradaris (literally defined as brotherhoods). Both bradaris contain children born to consanguineous (first cousin) and nonconsanguineous marriages. DNA samples have been supplied by Dr Subaib Ahmed of the Armed Forces Institute of Pathology in Rawalpindi, for a total of 91 individuals. The specific allele frequencies, and levels of homozygosity of each bradari, were determined using twenty fluorescence-labelled microsatellite markers for chromosomes 13 and 15. The project has studied the inheritance of microsatellite alleles across four chromosomes within two multi-generation families from a highly endogamous community, the Sankethi of Karnataka, India. The two families each have members from four generations participating in the study. Finger prick blood samples and detailed pedigree information were collected from subjects in India. DNA was extracted from the blood spot samples using a phenol-chloroform technique, and analysed by spectrophotometry to determine DNA concentrations. Fluorescent markers from chromosomes 15, 16, 17 & 18 were amplified using polymerase chain reaction (PCR) techniques on a Perkin Elmer 96 well thermocycler. The PCR products …

The study was conducted to determine the effects of lowered genetic variance in the Western mosquitofish (Gambusia affinis) on the physiological parameters of critical thermal maximum and minimum pH. Mosquitofish were obtained from two small central Illinois ponds, Lost Pond and Shadow Pond, by seines and dip nets. Once caught, fish were returned to the laboratory and acclimated for a minimum of five days at a temperature of 27°C and a pH of 8.25. At the beginning of each experimental run, fish were subjected to the physiological experiments for determination of critical thermal maximum and minimum pH, and then measured …
