Harmful algal blooms (HABs) are a serious environmental problem globally. The ability of cyanobacteria, one of the major causative agents of HABs, to grow in heavy metal polluted areas is proving a challenge to environmental restoration initiatives. Some cyanobacteria secrete toxins, such as microcystin, that are potentially dangerous to animals and humans. In this study, the physiology of a cyanobacterium was assessed to nickel chloride exposure. Cell growths were monitored throughout the study with various nickel chloride concentrations (0, 10, 25 or 50 mg/L). Morphological abnormalities were observed with microscopic image analyses. Tissue-associated bacterial community profiles generated using a nested polymerase chain reaction–denaturing gradient gel electrophoresis (DGGE) approach and culture-dependent and culture-independent isolation techniques were compared. Oyster samples were collected from 2 harvest areas along the coast of Maine, in the United States. Profiles from both isolation strategies were evaluated using Sorensen’s index of similarity and cluster analysis of gel banding patterns. Cultureindependent profiles were further evaluated using the Shannon diversity index. In general, the culture-dependent strategy resulted in a greater number of bands within a profile. A common member of the intestinal microbiota in humans and animals is Escherichia coli. Based on the presence of virulence factors, E. coli can be potentially pathogenic. The focus of this study was to isolate E. coli from untreated surface waters (37 sites) in Illinois and Missouri and determine phenotypic and genotypic diversity among isolates. Water samples positive for fecal coliforms based on the Colisure® test were streaked directly onto Eosin Methylene Blue (EMB) agar (37°C) or transferred to EC broth (44.5°C). The aim of this work was to optimize the biodegradation of polyvinyl alcohol (PVA) containing actual textile wastewater for a sustainable treatment solution. The isolated microbial consortia of effective PVA degrader namely Candida Sp. and Pseudomonas Sp., which were responsible for symbiotic degradation of chemical oxidation demand (COD) and PVA from desizing wastewater. In the process optimization, the maximum aeration was essential to achieve a high degradation rate, where as stirring enhances further degradation and foam control. This study reports a user-friendly technique in the preparation of fungal inoculum intended for solid-state fermentation (SSF) – Cellophane Film Culture (CFC) technique. This technique uses cellophane film-overlaid agar plates to facilitate the separation of fungal biomass. The findings showed that inoculum of P. sajor-caju produced is viable, and it was confirmed by the presence of laccase enzyme activity in SSF of rice husk. The correlation between fungal dry and wet weights (r2 = 0.9329) provides an accurate estimation of fungal dry weight from its wet weight during inoculum preparation. Half of the microbial cells in the Earth’s oceans are found in sediments. Many of these cells are members of the Archaea, singlecelled prokaryotes in a domain of life separate from Bacteria and Eukaryota. However, most of these archaea lack cultured representatives, leaving their physiologies and placement on the tree of life uncertain. Here we show that the uncultured miscellaneous crenarchaeotal group (MCG) and marine benthic group-D (MBG-D) are among the most numerous archaea in the marine sub-sea floor. To assess the applicability of latex cell coatings as an ‘off-the-shelf’ biocatalyst, the effect of osmoprotectants, temperature, humidity and O2 on preservation of H2 production in Rhodopseudomonas palustris coatings was evaluated. Immediately following latex coating coalescence (24 h) and for up to 2 weeks of dry storage, rehydrated coatings containing different osmoprotectants displayed similar rates of H2 production. Beyond 2 weeks of storage, sorbitol-treated coatings lost all H2 production activity, whereas considerable H2 production was still detected in sucrose- and trehalose-stabilized coatings. A study was performed in Del Rey Lagoon, City of Los Angeles, to determine if the lagoon was as a source or sink for fecal indicator bacteria (FIB: total coliforms, Escherichia coli, enterococci) and to screen for the presence of other potentially pathogenic bacteria. The lagoon receives tidal flows from the adjacent Ballona Estuary whose water usually is contaminated with FIB originating from the highly urbanized Ballona Creek Watershed.
