Additive manufacturing of polymers is gaining momentum in health care industries by providing rapid 3D printing of customizable designs. Yet, little is explored about the cytotoxicity of leachable toxins that the 3D printing process introduced into the final product. We studied three printable materials, which have various mechanical properties and are widely used in stereolithography 3D printing. We evaluated the cytotoxicity of these materials through exposing two fibroblast cell lines (human and mouse derived) to the 3D-printed parts, using overlay indirect contact assays. Microscopic bioimaging is a useful approach to study cell-biomaterial interactions which are vital to the biomedical application of biomaterials. Through microscopic imaging, numerous cellular responses, such as proliferation, uptake, and death, can be recorded, characterized and analyzed.
In this thesis, I first provided basic introductions to the imaging techniques and analysis tools, especially those that are highly relevant to the studies of biomaterials and cell interactions. I also detailed the adaptation of these techniques and tools in the application of two specific research projects in biomaterials, with special focuses on the imaging and analysis.

Sterilization of tissue engineered scaffolds is an important regulatory issue and is at the heart of patient safety. With the introduction of new biomaterials and micro/nano structured scaffolds, it is critical that the mode of sterilization preserve these built-in features. Conventional sterilization methods are not optimal for engineered polymeric systems and hence alternate systems need to be identified and validated. PCL is polyester with a low melting point (heat labile), susceptible to hydrolysis and is popular in tissue engineering. Biomedical devices that function in-vivo offer a tremendous promise to improve the quality of life for many who suffer from disease and trauma. The most important consideration for these devices is that they interact with the physiological environment as designed without initiating a deleterious inflammatory response. ISO 10993 outlines the current international guideline for investigating the biocompatibility of such devices. Numerous groups report the use of ISO 10993 as the basis for their experimental evaluation of candidate materials for neuroprosthetics, as well as other biomedical devices, however most of these reports fail to completely comply with the standard.
