N-Succinyl-L,L-2,6-Diaminopimelic Acid Desuccinylase (DapE) is a bacterial enzyme located in the lysine biosynthetic pathway of all Gram-negative and most Gram-positive species of bacteria including the notorious ESKAPE pathogens, or pathogenic strains of bacteria that have developed significant levels of resistance to currently available antibiotics. As a means to combat this challenge of growing antimicrobial resistance, we have identified the bacterial enzyme DapE as a conserved, novel enzymate target within these resistant bacterial strains,as DapE is responsible for the production of m-DAP and lysine which are ultimately employed in bacterial cell wall synthesis. Cancer is one of the leading causes of death in the world with over 10 million people dying of the disease every year. Inorganic complexes supported by platinum metal centers are often used in cancer treatment. However, these complexes lack selectivity for tumor cells. As such, other alternatives to platinum have been explored. Ruthenium (Ru) complexes are one of the most promising candidates due to their ability to be used as photosensitizing drugs in photodynamic therapy. Exposing The Effects Of Reaction Cavity In Solid-Phase Reaction Kinetics, Michael Carr

Reactivity of molecular solids is governed by topochemical considerations, where minimizedmolecular movement dictates both product identity and kinetics. This Thesis explores the central role reaction cavity plays in the topochemical principle by using the highly varied cavities formed by the different facets of a tetracene crystal. The relative rates for ten different reaction cavities were measured via energy dispersive X-ray spectroscopy for the reaction of tetracene with vapors of maleic anhydride. Variance in cavities resulted in Diels-Alder reaction rates that differed by as much as a factor of 2.6. Effects were understood via molecular dynamic simulations which modeled the approach …

Mechanistic Investigations Of The Iron(Iii)-Catalyzed Carbonyl-Olefin Metathesis Reaction, Susan Phan

Iron(III)-catalyzed carbonyl-olefin ring-closing metathesis represents a new approach toward the assembly of molecules traditionally generated by olefinˆ’olefin metathesis or olefination. Herein, we report detailed synthetic, spectroscopic, kinetic, and computational studies to determine the mechanistic features imparted by iron(III), substrate, and temperature to the catalytic cycle. These data are consistent with an iron(III)-mediated asynchronous, concerted [2+2]-cycloaddition to form an intermediate oxetane as the turnover-limiting step. Fragmentation of the oxetane via Lewis acid-activation results in the formation of five- and six-membered unsaturated carbocycles.

Building And Optimizing A Spectroelectrochemical System For The Study Of Shewanella Putrefaciens, Jonathan Muscolino

Shewanellaceae are among the most widely studied electroactive microorganisms. This study reports on Shewanella putrefaciens under anaerobic conditions during applied potential while monitored by spectroscopy. The attachment of the microbe Shewanella putrefaciens onto an indium tin oxide electrode was found to occur after 2 hr of +0.2 V applied potential. Microbial detachment occurred with a 5 mV/s scan rate cyclic voltammogram from +0.2 V to -0.5 V to +0.2 V. This was confirmed through use of a spectroelectrochemical apparatus utilizing a standard 3 electrode setup with evanescent wave spectroscopy.

Computer simulations using molecular dynamics (MD) on classical molecular mechanical

(MM) interatomic potentials can provide valuable information and quantitative

predictions about these systems. In MD calculations binding free energies of ions to

host molecules can be studied if correct ion solvation free energies in aqueous solution

are obtained. However, no MM models exist that parametrize various Zn2+ and

transition metals consistently and consider both structural and thermodynamic data

simultaneously.

The first part of our work focused on MD free energy perturbation (FEP) simulations

to derive MM interaction parameters for Zn2+ and Mg2+ ions in aqueous
