The common assumption in regard to metabolic rate scaling with body size is the 3/4 law, which predicts a scaling exponent of 0.75 between log metabolic rate and log body size. Supporting evidence exists for this theory on a large, general scale. However, factors such as temperature, predation, and environment can cause the scaling exponent to deviate from 0.75. This paper takes a closer look at the effects of temperature and predation on metabolic rates via oxygen consumption in freshwater macroinvertebrate populations. CRISPR-Cas9 genome editing has been used successfully to knock out genes in model organisms such as zebrafish, turquoise killifish, and cichlid fish. CRISPR-Cas9 genome editing has not been verified in the annual killifish, Austrofundulus limnaeus. We hypothesize that targeted editing of the tyrosinase gene in embryos of A. limnaeus will lead to fish without the ability to produce black pigment. Embryos at the 1-cell stage were injected with a Cas9 cocktail containing a mix of guide RNA molecules that target the genomic sequence of the tyrosinase gene and either an mRNA coding for the Cas9 protein or Cas9 protein. …

Due to the alarming rates of freshwater fish extinctions, urgent action is needed to develop captive breeding programs for imperiled species and enhance existing practices to improve reproductive outcomes. Here, we investigated the effects of enrichment on gamete production, quality, and spawning coloration following hormone injection (i.e. carp pituitary, gonadotropin) in the endangered redside dace Clinostomus elongatus, a sexually dimorphic, presumably obligate nest parasite. C. elongatus were reared in either a non-enriched environment (i.e. barren) or an enriched environment (i.e. substrate, plants, and spawning nest-building hosts) for 1 yr prior to hormone induction. Seagrass beds support high biodiversity and animal abundance, serve as feeding grounds for a variety of animals, offer shelter from predation, and act as a nursery habitat for juveniles. The species composition of seagrass beds can impact their use as habitat by animals. Two common species of seagrass in the Gulf of Mexico are Ruppia maritima (widgeon grass) and Halodule wrightii (shoal grass). The swordtail, Xiphophorus hellerii, is a freshwater fish species native to Mexico and Central America and is commonly used in the tropical aquarium trade. Swordtails have been shown to have limited survival in 6 ‰ salinity (Nanda et al., 2016), suggesting they may have a greater salinity tolerance than previously expected. Using a gradual acclimation method, we examined the salinity tolerance and swim performance of this species. Freshwater-reared female swordtails were housed in 29-gallon aquaria. Freshwater fishes are extraordinarily diverse, considering their available habitats represent a tiny proportion of the earth’s surface. Rivers connect heterogeneous habitats in a linear form and provide excellent simplified models to understand how aquatic biodiversity evolves. In particular, the lower Congo River (LCR) in west Central Africa consists of a dynamic hydroscape exhibiting extraordinary aquatic biodiversity, endemicity, and morphological and ecological specialization. This system is thus an excellent natural laboratory for understanding complex speciation and population diversification processes. White sturgeon (Acipenser transmontanus) in the Lower Fraser River are the focus of a catch-and-release angling fishery in British Columbia, Canada. However, the lower region of the catch area includes areas where tidal waters invade, and the consequence of salinity levels on recovery from an angling challenge are not characterized in sturgeon, despite theoretical implications of its import. We acclimated white sturgeon to various salinities (0, 10 and 20 (parts per thousand)) to investigate the effects of acclimation on recovery from stimulated angling stress that was induced through manual chasing. Amphibians are important members of freshwater communities and are susceptible to negative effects of climate change and invasion. Furthermore, both climate change and invasion can influence density of amphibian larvae at the microhabitat scale because of larval clustering. This research provides insight on the effects that polyunsaturated aldehydes (PUAs) have on model species zebrafish heart rate and mortality. Zebrafish were used as an analog for forage fish, which are of both ecological and economic importance. Forage fish often interact with diatoms, some species of which have been known to emit allelopathic chemicals, PUAs. These chemicals can have a deleterious effect on invertebrates that cohabitate with or consume diatoms, but their effects on vertebrates was unknown. Analogous to the syrinx in birds and larynx in mammals, the swim bladder is a major sound-producing organ in fishes. It also functions in hearing and is responsible for most returns from sonar. Historically, the swim bladder has been modeled as an underwater resonant bubble, whereas we provide evidence for a forced-response model in which viscous damping of the bladder wall inhibits resonance of the internal bubble. Further experimental work on the shape, wall structure, and mechanical behavior of the swim bladder is required.

Fishes are surrounded by various types of stimuli in their habitats which provide important information about their environments. Here, I investigated how various types of stimuli can affect the behaviour and physiology of two freshwater species: the invasive round goby (Neogobius melanostomus) and the black bullhead (Ameiurus melas). In chapter 2, I tested for a synergistic response in the round goby to multimodal stimuli, by presenting acoustic, visual, and olfactory stimuli separately and simultaneously. The sea lamprey is a basal vertebrate, and the oldest extant species with taste buds. The objective of this thesis was to study the chemosensory responses from the sea lamprey’s pharynx, which is located caudal to oral cavity and medial to seven lateral brachiopores. The pharynx contains taste buds that detect and respond to tastants to inform lampreys about gustatory factors in the environment. The location of these taste buds along the lateral surface of the pharynx was first determined using scanning electron microscopy. Environmental DNA (eDNA) sampling provides a method for assessing fish communities that has potential as a supplement to traditional sampling methods due to its ability to save time as well as its non-invasive nature. Lake sturgeon (Acipenser fulvescens) are endangered in the Laurentian Great Lakes with increasing binational efforts to establish spawning grounds to aid restoration. While SCUBA surveys can document spawning activity, these are labour-intensive and may disrupt spawning. We used passive acoustic monitoring to quantify spawning sounds of lake sturgeon as a first step to developing remote sensing of sturgeon spawning grounds. Acipenser sp. are known to make a variety of sounds including, “thunders” (aka drums), which have been documented in A. fulvescens during spawning. We quantified drums from a known spawning bed. The accurate determination of age for fish is a vital part of both populationmanagement and ichthyological research. However, the methods which are primarily employed to age fish can be difficult, time consuming, inaccurate, or some combination thereof. Most ageing is currently done via hard parts (such as otoliths, vertebrae, or scales) which are read in a manner similar to tree rings with markings corresponding to known or presumed periods of time. The Sangamon River flows across central Illinois and is mostly free flowing except in the city of Decatur where it is impounded to form Lake Decatur. The Sanitary District of Decatur (SDD) is located approximately three miles downstream of the Lake Decatur dam. For a significant portion of the year, the stretch of the Sangamon River downstream of SDD is dominated by effluent due to limited discharge from the Lake Decatur dam. Fisheries populations and their ecosystems are negatively impacted by both chronic and acute inputs of pollutants, including oil spills such as the Deepwater Horizon platform blowout in 2010 in the Gulf of Mexico (GoM). After Deepwater Horizon, toxicological studies of demersal fishes of the northern GoM were undertaken to characterize impacts and to establish baseline contaminant levels in the aftermath of the spill. Although the hypoxia-inducible transcription factor (HIF) orchestrates molecular responses to low oxygen, the amount of HIF-1α protein expressed during hypoxia varies among species, individuals, and tissues. This study measured HIF-1α protein levels along with several physiological and behavioral variables in the estuarine fish Fundulus grandis under normoxia (> 7 mg l-1 dissolved oxygen) and hypoxia (1 mg l-1 dissolved oxygen). Fish under hypoxic conditions had higher tissue levels of HIF-1α, hematocrit, blood glucose, blood lactate, frequency of aquatic surface respiration (ASR), and lower activity than normoxic controls. Silver (Hypopthalmichthys molitrix) and bighead (H. nobilis) carp, are invasive species that have negative impacts upon ecosystems. H. molitrix is known to jump completely out of the water in response to broadband sounds, however, this is not observed in H. nobilis. Preliminary experiments reveal that sounds can be used to modify the behavior of carps. Thus, understanding the hearing abilities of these species is important in order to design appropriate acoustical deterrents. Network representation provides a natural framework for the study of real world complex systems. From social networks and animal groups to interneuronal communications and power grid systems, complex patterns of interaction can be captured and modeled using networks in a simple mathematical form. In many cases, however, a faithful network representation of the system is not readily available. For this reason, network reconstruction has become a growing topic of interest in recent years, the goal of which is to discover the hidden interaction patterns among individuals by fitting input-output data from multiple experiments to candidate network topologies.
