Human adenovirus type 64 (Ad64) is a causative agent of contagious viral pink eye. We do not understand why Ad64 causes eye infections. One likely reason is the presence of a receptor molecule on the exposed surface of eye cells, but not other cells. Ad64 binds to a protein called CD46 on the cell surface. We aim to visualize how CD46 binds to the surface of Ad64. I mixed the extracellular portion of CD46 with purified Ad64 virus, then visualized it using negative stain transmission electron microscopy. The soil bacterium Agrobacterium tumefaciens causes tumors in plants through interkingdom gene transfer. This transfer is initiated upon a wounding event that results in the release of plant signaling factors such as phenols and sugars that are recognized by protein machinery in the periplasm and inner membrane of A. tumefaciens. The histidine kinase protein VirA, in combination with the periplasmic protein ChvE, recognizes these signals and initiates induction of virulence genes via a signaling pathway, culminating with the insertion of tumor-inducing T-DNA into the wounded plant cells. Due to advances in high-throughput parallel sequencing, researchers have conducted novel studies exploring relationships between microbiome compositions and different aspects of organism health. Some of these studies have shown that the gut microbiome of rodent models has effects on organism health and behavior and that infection with pathogens and the composition of the skin microbiome are linked to changes in gut microbiome composition. Mitochondrial homeostasis requires a dynamic balance of fission and fusion. The actin cytoskeleton promotes fission, and we found that the mitochondrially localized myosin, myosin 19 (Myo19), is integral to this process. Myo19 knockdown induced mitochondrial elongation, whereas Myo19 overexpression induced fragmentation. This mitochondrial fragmentation was blocked by a Myo19 mutation predicted to inhibit ATPase activity and strong actin binding but not by mutations predicted to affect the working stroke of the motor that preserve ATPase activity. Super-resolution imaging indicated a dispersed localization of Myo19 on mitochondria, which we found to be dependent on metaxins. These observations suggest that Myo19 acts …

Halogen bonding is a noncovalent interaction that continues to garner interest among the scientific community. Investigation of halogen bonds in biological contexts typically revolves around rational drug design for developing therapeutics. However, halogen bonding may be occurring naturally in our body every day. Thyroid hormone and its regulating enzyme, iodothyronine deiodinase, show promising results for a halogen bonding interaction that happens during catalysis. Prior work has examined the interaction between the iodine of thyroid hormone and the selenium of iodothyronine deiodinase. But in recent years, the concerns over indoor air pollution has increased. People can spend up to 90% of their time in indoor environments, especially their homes, even more so since the beginning of the COVID-19 pandemic. We describe the internal oral morphology and chondrocranial anatomy for Boana crepitans tadpoles, and compare them with available descriptions for other species in the subfamily Cophomantinae. Among species of the Boana faber group, the chondrocranial anatomy has been reported only for one species internal oral morphology and cranial anatomy are similar to other described species of Boana and Cophomantinae. B. crepitans lacks unique features in the oral cavity and chondrocranium that would distinguish it from other congeneric species. In this project, we aim to functionally test the contribution that individual Cdx binding sites have in hox gene regulation. We will achieve this by individually blocking Cdx binding sites using CRISPR/dCas9 in the zebrafish and then analyzing changes in the time, distribution, and levels of hox gene transcription.

These diseases can be caused by the burning of biomass (plastic, wood, rubber, tobacco, etc.) for light or heat. Acrolein is a common indoor and outdoor pollutant from tobacco smoke or organic combustion. The Leptodactylus latrans species group currently comprises eight medium- to large-sized frog species with a convoluted taxonomic history, particularly related to the specific limits of the L. latrans complex, and the species pair Leptodactylus chaquensisLeptodactylus macrosternum. Their homogeneous external morphology and continental geographic distribution in South America have posed severe limitations to a comprehensive review, such that taxonomic consensus and species limits remain uncertain. Invasive species are ideal systems for testing geographical differences in performance traits and measuring evolutionary responses as a species spreads across divergent climates and habitats. The European gypsy moth, Lymantria dispar dispar L. (Lepidoptera: Erebidae), is a generalist forest defoliator introduced to Medford, Massachusetts, USA in 1869. The invasion front extends from Minnesota to North Carolina and the ability of this species to adapt to local climate may contribute to its continuing spread. The genus Edalorhina consists of two species of small forest-floor frogs inhabiting the Amazon basin. The tadpole of Edalorhina perezi, the most widely distributed species, was previously described based on a single and early stage (Gosner 25) individual. Herein, we provide a description of the tadpole in Gosner stages 35–36 including internal morphology data (i.e., buccopharyngeal cavity and larval skeleton) based on samples from two populations from Ecuador. 8-oxo-2’-deoxyguanosine (OdG) is a prominent DNA lesion that can direct the incorporation of dCTP or dATP during replication. As the latter reaction can lead to mutation, the ratio of dCTP/dATP incorporation can significantly affect the mutagenic potential of OdG. Previous work with the A-family polymerase BF and seven analogues of OdG identified a major groove amino acid, Ile716, which likely influences the dCTP/dATP incorporation ratio opposite OdG. To further probe the importance of this amino acid, dCTP and dATP incorporations opposite the same seven analogues were tested with two BF mutants, I716M and I716A. The Allee threshold, the critical population density separating growth from decline in populations experiencing strong Allee effects, can vary over space and time but few empirical studies have examined this variation. A lack of geographically extensive, long-term studies on low density population dynamics makes studying variability in Allee effects difficult. We used North American gypsy moth population data from 1996-2016 to quantify Allee thresholds in 11 regions of the invasion front. Allee thresholds spanned a continuum from being undetectable due to strong population growth at all densities, to being unmeasurable because populations declined across all densities. Tsetse flies (Diptera: Glossinidae) house a taxonomically diverse microbiota that includes environmentally acquired bacteria, maternally transmitted symbiotic bacteria, and pathogenic African trypanosomes. Sodalis glossinidius, which is a facultative symbiont that resides intra and extracellularly within multiple tsetse tissues, has been implicated as a mediator of trypanosome infection establishment in the fly’s gut. Tsetse’s gut-associated population of Sodalis are subjected to marked temperature fluctuations each time their ectothermic fly host imbibes vertebrate blood. Leptodactylus guianensis is reported for the first time for Colombia along with a distributional map of the species. The species is the fifth species of the L. latransspecies group documented in the country. Males possess a single, conical and slightly chisel-shaped thumb spine; only three species in the group have a single thumb spine. In collections, specimens of L. guianensis are misidentified as L. macrosternum, L. latrans or L. bolivianus. We provide morphological, colouration and habitat descriptions for where the species occurs in Colombia.

Retinoic acid is a known morphogen in regulating animal growth and development. Planaria are a key model system for regeneration and their eyes are a morphological marker of anterior differentiation. We explored the requirement for retinoic acid signaling in the regeneration of body parts in the planaria S. mediterranea using an inhibitor of retinoic acid synthesis, diethylaminobenzaldehyde (DEAB). Whole planaria, soaked in DEAB for three days prior to and five days following amputation, produced trunk and tail fragments with defective anterior regeneration. Following regeneration, up to 80% of posterior fragments developed abnormal eyes. The abnormalities included animals without eyes, with …

Chiasmocleis is the most species-rich genus of Neotropical microhylids. Herein, we provide the first comprehensive multilocus phylogeny for the genus, including all but 3 of the 34 recognized species and multiple individuals per species. We discuss cryptic speciation, species discovery, patterns of morphological evolution, and provide a historical biogeographic analysis to account for the current distribution of the genus. Diversification of Chiasmocleis from other New World microhylids began during the Eocene, app. 40 mya, in forested areas, and current diversity seems to be a product of recurrent connections between the Atlantic Forest and Amazonia. The family Rivulidae is the fourth most diverse clade of Neotropical fishes. Together with some genera of the related African family Nothobranchiidae, many rivulids exhibit a characteristic annual life cycle, with diapausing eggs and delayed embryonic development, which allows them to survive in the challenging seasonal ponds that they inhabit. Rivulidae also includes two species known as the only the self-fertilizing vertebrates and some species with internal fertilization. The first goal of this article is to review the systematics of the family considering phylogenetic relationships and synapomorphies of subfamilial clades, thus unifying information that is dispersed throughout the literature. The Neotropical genus Leptodactylus is currently represented by three species in the West Indies (Leptodactylus albilabris, Leptodactylus fallax and Leptodactylus validus). Based on morphological, acoustic and molecular evidence, we document the presence of a fourth species in the Caribbean region, Leptodactylus fragilis (Brocchi, 1877). The species was found at two localities in western Cuba, and molecular data suggest a northern South American origin, possibly Venezuela, for these populations. We discuss the potential invasive status of L. fragilis, based on its known distribution, relative abundance, behaviour and possible impacts on native species of Cuban amphibians.

For organisms with complex life cycles, conditions experienced during early life stages may constrain later growth and survival. Conversely, compensatory mechanisms may attenuate negative effects from early life stages. We used the spotted salamander, Ambystoma maculatum, to test how aquatic larval density and terrestrial moisture influence juvenile growth, food intake, evaporative water loss and water reuptake rates, and corticosterone levels. We conducted an outdoor mesocosm experiment to manipulate larval density and transferred metamorphosed salamanders into low and high terrestrial moisture treatments in laboratory terrariums. In the context of a new science of heroism this article presents a brief history of the presence and study of heroism, and an outline of the key characteristics of the modern-day movement of heroism. The potentially wide-reaching impacts of the science of heroism are briefly discussed. The hypothesis of human beings as “hero organisms” is presented, before embarking on a discussion on what it means for each of us to be on our own hero’s journey.

Traits that promote the maintenance of body temperatures within an optimal range provide advantages to ectothermic species. Pigmentation plasticity is found in many insects and enhances thermoregulatory potential as increased melanization can result in greater heat retention. The thermal melanism hypothesis predicts that species with developmental plasticity will have darker pigmentation in colder environments, which can be an important adaptation for temperate species experiencing seasonal variation in climate. The harlequin bug (Murgantia histrionica, Hemiptera: Pentatomidae, Hahn 1834) is a widespread invasive crop pest with variable patterning where developmental plasticity in melanization could affect performance.
