Genetic Mechanisms Underlying Maladaptation In Specialized Species Interactions, Nitin Ravikanthachari

Adaptation has been used as the framework to understand the power of natural selection in structuring biodiversity and driving biological interactions. However, recent examples have indicated that many organisms are maladapted to their local conditions. Maladaptation can result from various causes including rapid environmental change, novel environments, interplay of evolutionary forces (viz: gene flow-selection balance) and anthropogenic disturbance.

To dissect the genetic mechanisms underlying maladaptation, I used the interaction between Pieris macdunnoughii (Remington) (Lepidoptera: Pieridae) which lays eggs on an invasive mustard Thlaspi arvense (L.) (Brassicaceae), which is lethal to the larvae.

Autism Spectrum Disorders (ASD) are highly heritable neurodevelopmental disorders characterized by social and verbal impairments as well as repetitive behaviors. Large human genetic studies showed that genes encoding chromatin and transcriptional regulatory proteins are among the most commonly mutated genes in ASD. Absent, Small, or Homeotic 1-like (ASH1L) is a histone methyltransferase that has previously been identified as a major genetic risk factor for ASD. ASH1L di-methylates Histone H3 on Lysine 36 (H3K36me2) and tri-methylates Histone H3 on Lysine 4 (H3K4me3), which mainly coincides with transcriptional activation. In Arabidopsis thaliana, Enhanced Disease Susceptibility1 (EDS1) acts as an indispensable hub in plant immunity and is mainly responsible for plant basal defense, systemic acquired resistance (SAR), increase of endogenous content of salicylic acid (SA) and effector-triggered immunity (ETI). Epigenetic regulators play crucial roles in multiple important cellular processes. As one catalytic core of Polycomb Repressive Complex (PRC2), CURLY LEAF (CLF/SDG1) protein promotes H3K27me3 deposition at specific loci for genes repression. Bobcats (Lynx rufus) are an ecologically and genetically diverse species with a large contiguous range throughout North America. The species not only has a wide array of phenotypic variation compared to other mammals, but shows marked adaptability across ecozones with differing ecological influences. It is these various selective pressures in distinctive parts of the continent that have likely led to localized adaptations within the bobcat metapopulations. Nanotechnology takes advantage of cellular biology’s natural nanoscale operations by interacting with biomolecules differently than soluble or bulk materials, often altering normal cellular processes such as metabolism or growth. To gain a better understanding of how copper nanoparticles hybridized on cellulose fibers called carboxymethyl cellulose (CMC) affected growth of Saccharomyces cerevisiae, the mechanisms of toxicity were explored. Multiple methodologies covering genetics, proteomics, metallomics, and metabolomics were used during this investigation. The work that lead to this dissertation discovered that these cellulosic copper nanoparticles had a unique toxicity compared to copper. Sporulation is a process in which some bacteria divide asymmetrically to form tough protective endospores, which help them to survive in a hazardous environment for a quite long time. The factors which can trigger this process are diverse. Heat, radiation, chemicals and lacking of nutrition can all lead to the formation of endospores. This phenomenon will lead to low productivity during industrial production. However, the sporulation mechanism in a spore-forming bacterium, Clostridium theromcellum, is still unclear. The C.elegans germline offers an ideal system to study posttranscriptional regulation of gene expression as it is a major mechanism through which the control over gene expression is achieved. GLD-1 (defective in GermLine Development) is a maxi-KH motif containing RNA binding protein that controls various aspects of germline development from decision over germcell proliferation vs. meiotic entry to the production of mature gametes suggesting that GLD-1 likely controls many mRNA targets.

Cave systems and their unique biota are widely viewed as highly endangered, yet very little is known about basic life history, ecology, distributions, habitat requirements, and evolutionary relationships of subterranean species. The crux of the problem in cave studies is the assumption that traditionally defined morpho-species represent distinct evolutionary lineages. Convergence is exhibited in the morphologies of many animal groups, vertebrate and invertebrate, which leads to confusion in diagnosing species' boundaries, geographic distributions, gene flow routes, and imperilment. This dissertation research includes phylogeographic analyses of freshwater cave-dwelling crayfishes in the Southern Appalachians, a global hotspot of subterranean biodiversity. By examining …

The blue catfish, Ictalurus furcatus (Ictaluridae), is ranked among the most invasive, nonnative species of concern in the Chesapeake Bay watershed. This species, intentionally introduced to three major tributaries and a number of impoundments between 1974 and 1989 for sport fishing, has spread into three additional tributaries. Using samples from the introduced tributary populations as a baseline, we evaluated microsatellite genetic variation in light of demographic and ecological data to elucidate the potential sources of the invasive I. furcatus populations. The pathogenesis of autism has proven difficult to characterize. However, in many recent studies, it is suggested that the onset of this disorder is the result of multiple etiological factors, which include genetic, immunologic, and viral elements.

Possible immunological influences found in subpopulations of patients with autism include decreased lymphocyte responsiveness, reduced natural killer cell activity, abnormal response to rubella vaccine, abnormal immune response to brain tissue, and decreased plasma levels of the fourth component of complement(C4). These aberrations and others imply a possible autoimmune mechanism in some cases for the development of autism.
