Lake Mead is a deep, subtropical, moderately productive, desert impoundment with a negative heterograde oxygen profile occurring during; the summer stratification. investigations of the Boulder Basin of Lake Mead by the University of Nevada were initiated in November 1971. The primary objective of the study was to determine what effects industrial and sewage effluent from the Las Vegas metropolitan area, discharged into Las Vegas Bay, have had on the water quality and limnological conditions of Boulder Basin. Data from the 1975-76 period are presented in detail, with earlier data included in the summaries and discussions.

The zero‐frequency shear wave dip appearing in the depolarized Rayleigh spectrum in benzonitrile has been studied as a function of concentration and temperature. The solution study was carried out at constant viscosity equal to the viscosity of liquid benzonitrile at each temperature. The result indicates that the presence of shear wave fine structure does not depend on the collective orientational fluctuations. The orientational and vibrational relaxation times of benzonitrile were measured at various concentrations and temperatures. The orientational relaxation times show no concentration dependence at any temperature, suggesting that the pair correlation is negligible at all concentrations. The orientational relaxation …

Human placental alkaline phosphatase was purified by butanol extraction, methanol precipitation and chromatography on DEAE-cellulose. The pH optimum was found to be 10.9. The enzyme was competitively inhibited by di-ethyl-p-nitrobenzyl-phosphonate and copper sulfate. An increase in enzymatic activity occurred in the presence of the phosphate ion (HPO4) and ATP.

The enzyme was crystallized from a 40% saturated solution of ammonium sulfate and shown to be homogeneous by sedimentation analysis. Homogeneity of the enzyme was also demonstrated using poly-acrylamide-gel-electrophoresis. At a pH of 7.5% the electrophoretic pattern yielded a single band and showed no significant contamination.

The Relationship Of Reduced Levels Of Atp To Inhibited Chondrogenesis In The Production Of Limb Deformity Induced By 6-Aminonicotinamide, Val C. Sheffield

6-aminonicotinamide (6-AN), an analogue and antagonist of nicotinamide, impairs cartilage formation and results in shortening of the limb when administered to chick embryos. 6-AN forms an abnormal NAD analogue (6-ANAD), which inhibits the activity of NAD-dependent enzymes associated with the production of ATP. The mechanism of action of 6-AN was studied by measuring the biosynthesis of protein and DNA, and the sulfation and glycosylation of chondroitin sulfate in control and 6-AN treated cartilage from 8-day chick embryos. The cartilage was also assayed for ATP levels. Incorporation of Na235SO4 was inhibited by 6 h treatment with 10 μg/ml 6-AN, whereas 3H-thymidine …

The rotational and vibrational relaxation times of acetonitrile–carbon tetrachloride solutions were investigated as a function of concentration, viscosity, and temperature using depolarized Rayleigh and Raman scattering. Using a Fabry‐Perot interferometer and single frequency laser source, we have shown that reliable results for the single particle orientational correlation times (τs) for CH3CN can be obtained by carrying out a concentration dependent depolarized Rayleigh scattering study. Raman scattering was shown to yield inconsistent results for τs in CH3CN. At constant viscosity, it was found that the Rayleigh scatteringrelaxation time (τRay) of CH3 …

Experiments were undertaken to identify and characterize control mechanisms in tobacco leaf tissue which decrease the relative contribution of photorespiratory CO2 release and thereby increase net photosynthetic CO2 fixation. A number of metabolites were supplied to illuminated leaf discs and their effect on the inhibition of glycolate synthesis was measured. Glycolate accumulation, in the presence of α-hydroxy-2-pyridinemethanesulfonic acid, was inhibited in leaf discs previously floated on 30 mM solutions of either L-glutamate, L-aspartate, phospho-enolpyruvate, or glyoxylate. The effect of glutamate on glycolate synthesis, which was investigated in detail, was concentration- and time-dependent. Glycolate synthesis was inhibited about 40% by treating …

Protoplasts were isolated from tobacco and barley leaves in sucrose or mannitol using commercially available cellulases and macerozymes. Barley growth and protoplast isolation and purification conditions were optimized so that protoplasts were obtained in high yields free of unwanted debris and organelles. A technique for processing barley and tobacco protoplasts for examination by scanning electron microscopy was developed in which protoplasts seem to have maintained their structural integrity. Barley and tobacco protoplasts took up 3H-B. subtilis DNA, 125I-B. subtilis DNA or 125I-M. luteus DNA as a linear function of time (0-6 hr) and DNA concentration (0-200 μg/ml). Up to 16 …

The 5'-terminus of poliovirus polyribosomal RNA is pUp. A candidate for the 5'-terminus of poliovirion RNA was recovered as a compound migrating toward the cathode when 32P-labeled virion RNA was completely digested with ribonucleases T1, T2 and A and analyzed by paper ionophoresis at pH 3.5. Treatment with proteinase K reversed its direction of migration, indicating the presence of protein. Treatment with venom phosphodiesterase liberated all of the radioactivity as pUp, suggesting that poliovirion RNA has a protein-pUp 5'-terminus. Treatment of virion RNA with T1 ribonuclease alone generated a proteinase K-sensitive oligoribonucleotide. Analysis of the oligoribonucleotide using ribonucleases A and …

Intermolecular Potential Surfaces From Electron Gas Methods. Ii. Angle And Distance Dependence Of The A’ And A” Ar–No(X^2ii) Interactions, Glen C. Nielson, Gregory A. Parker, Russell T. Pack

Angle dependent intermolecular potential energy surfaces for the two states (2A’ and 2A”) that arise from the interaction of ground (X^2II) state NO with Ar are calculated using the electron gas model to obtain the short range interactions. The average and difference of the two interaction energies are fit to analytic forms convenient for use in scattering calculations and joined smoothly onto the long range van der Waals potential previously determined. The results, which appear to be of useful accuracy, and the applicability of the electron gas model to such open shell–closed shell interactions are discussed.

Naturalists have often marveled at the ability of some air-breathing vertebrates to remain underwater for long periods of time (that is, "long" from man's reckoning). Seals, penguins, porpoises, and whales are all noted fro their ability to use oxygen stored in various ways to permit them to "stay under" for many minutes.

Protein subunit structure in the nuclear envelope has not been previously described. Covalent crosslinking of polypeptides provides a method for studying the subunit structure of protein-rich systems. In this study, several polypeptide crosslinking methods have been used to determine associations among polypeptides in isolated chicken erythrocyte nuclear envelope. Interchain associations that exist in the isolated membrane were fixed by crosslinking the polypeptides in the isolated nuclear envelope. Crosslinked and uncrosslinked polypeptides were resolved by electrophoresis in a dissociating detergent system. Each agent caused distinctive alterations in the gel pattern.
