Increased Hydrogen Production By Genetic Engineering Of Escherichia Coli, Zhanmin Fan, Ling Yuan, Ranjini Chatterjee

Escherichia coli is capable of producing hydrogen under anaerobic growth conditions. Formate is converted to hydrogen in the fermenting cell by the formate hydrogenlyase enzyme system. The specific hydrogen yield from glucose was improved by the modification of transcriptional regulators and metabolic enzymes involved in the dissimilation of pyruvate and formate. The engineered E. coli strains ZF1 (ΔfocA; disrupted in a formate transporter gene) and ZF3 (ΔnarL; disrupted in a global transcriptional regulator gene) produced 14.9, and 14.4 µmols of hydrogen/mg of dry cell weight, respectively, compared to 9.8 µmols of hydrogen/mg of dry cell weight …

The objective of this research was to determine the effects of conventional inputs on soil chemical and biological properties compared to organic systems in a four year vegetable rotation. Tillage and cover crops were the same in all treatments to avoid confounding factors often present in similar research. Additional experiments investigated plant gene expression in organic and conventional management systems and in soils with decreased microbial diversity. Experimental plots were prepared in the spring of 2004; four replications of three management treatments, organic, low-input and conventional, were arranged in a randomized complete block design. This thesis evaluates the effectiveness of using alcohol sales as an indicator of development or lag associated with development in Kentucky counties using summary statistics, shift-share analysis and an econometric model. The summary statistics are used to evaluate possible lines of causality and the Shift-share analysis and econometric model deal the specific characteristics of the county that are assumed to be influenced by the sale of alcohol. Limitations to the county level data were limiting to the time period evaluated in this thesis. Obesity, the second leading cause of preventable death in the U.S., and related health problems increase people’s concerns about healthy food consumption. The increased prevalence of obesity is a major concern of societies both in developed and developing countries. Nutrition label usage has been increasing due to the link between diet and health. As intra-industry trade increases in U.S. agricultural and food processing industries, the historical agricultural trade surplus is tightening. In efforts to maintain the trade surplus a focus has shifted towards the promotion of agricultural and processed food exports among small and medium sized firms. This study intends to identify and evaluate the potential for exports among small to medium sized agricultural and food processing firms in Kentucky through a collection of survey data. Food safety events receive substantial media coverage and can create devastating economics losses for agribusiness firms. It is unclear what factors influence consumers’ purchasing decisions before or after a food safety event occurs. The objectives of this study is to identify these factors that influence purchasing decisions, determine how consumers respond to hypothetical food safety events, and compare these findings across different products and geographical regions. The data for this research was obtained from two surveys. One survey concerned fresh produce while the second focused on meat products. This study was one of a cluster of studies that originated via requests for proposals from the NIOSH National Agricultural Tractor Safety Initiative. The present study design consisted of several steps: (1) formation of an advisory group, (2) development and testing of a standard paper self-responding survey instrument, (3) sample selection of farm equipment dealers, (4) administration of the survey, (5) assessment and analysis of the survey, and (6) in-person response panel of dealers (n = 80) to review results of the questionnaire for further definition and sharpening of the recommendations from the survey.
